Il fagosoma si fonde con un numero variabile di
lisosomi dando il fagolisosoma e il patogeno è
esposto all’azione di un largo pannello di enzimi litici.
La fagocitosi porta alla produzione di metaboliti
reattivi dell’ossigeno che esplicano azione
battericida. Fagociti attivati da citochine
proinfiammatorie esprimono una forma inducibile di
monossido di azoto sintetasi caratterizzata dalla
capacità di produrre grandi quantità di monossido di
azoto, dotato anche esso di attività battericida.
4) Digestione
Degradazione nel fagolisosoma di macromolecole di
origine esogena ad opera di varie attività
enzimatiche. Nel caso di macrofagi segue la
presentazione antigenica nel contesto MHC classe II
e l’avvio di una risposta specifica
Meccanismi di “escape” della fagocitosi
1) Riconoscimento
Alcuni batteri operano meccanismi di “masking” e
prevengono in questo modo la fagocitosi ad opera

di PMN (ex: Yersinia enterocolitica, Salmonella
spp., Listeria spp.)

Once they enter tissues, monocytes cells mature and become macrophages. Macrophages may assume different morphologic
forms after activation by external stimuli, such as microbes. Some develop abundant cytoplasm and are called epithelioid
cells because of their resemblance to epithelial cells of the skin.
Macrophages in different tissues have been given special names to designate specific locations. For instance, in the central
nervous system, they are called microglial cells; when lining the vascular sinusoids of the liver, they are called Kupffer
cells; in pulmonary airways, they are called alveolar macrophages; and multinucleate phagocytes in bone are called
osteoclasts.

Le cellule NK esprimono il CD16, un recettore per la regione Fc delle IgG (v. Struttura
degli anticorpi, più avanti), e possono utilizzare questo recettore per mediare un altro tipo
di lisi cellulare che non presenta restrizione MHC. La citotossicità cellulo-mediata
anticorpo-dipendente (Ab-Dependent Cell-mediated Cytotoxicity, ADCC) dipende dalla
presenza di Ac che riconoscono una cellula bersaglio (la sua specificità è pertanto dovuta
alla specificità dell'Ac). In seguito al legame con il suo Ag, la regione Fc dell'Ac viene
esposta e si lega poi al suo recettore sulla cellula NK per formare un ponte molecolare.
Una volta formato il ponte, alla cellula bersaglio viene trasmesso un segnale litico ancora
non del tutto compreso, il quale ne determina la morte.
