La localizzazione del linguaggio articolato Tema centrale del dibattito scientifico a partire dalla prima met dell 800. Nello studio scientifico del linguaggio umano le afasie costituiscono una fonte inesauribile di conoscenze. Afasia = disturbo acquisito del linguaggio proposizionale, mentre il linguaggio automatico (filastrocche, canzoncine, contare) e la gestualit sono relativamente preservati (a meno che la lesione interessi anche aree somatomotorie).

Afasia di Broca
Afasia descritta da Broca in un primo paziente, Monsieur Leborgne, detto Tan , affetto da epilessia fin da bambino e che dallet di 21 anni aveva perso la capacit di parlare ( afemia ) ma era in grado di comprendere il linguaggio e di esprimersi mediante gesti. In seguito aveva sviluppato una emiplegia. Broca ebbe occasione di vedere il paziente solo una settimana prima della morte, a causa di un gangrena della gamba. Il giorno dopo la morte, il 18 aprile 1861, Broca present i risultati dell analisi autoptica del cervello di Leborgne alla seduta della Societ di Antropologia di Parigi (da lui fondata), suggerendo che la causa dellafasia era dovuta ad una degenerazione del piede della terza circonvoluzione frontale di sinistra (oggi nota come circonvoluzione di Broca ) In seguito Broca osserva altri pazienti con lo stesso tipo di afasia e si rende conto che tutti hanno la stessa lesione del lobo frontale sinistro.

For listening to and understanding spoken words, the sounds of the words are sent through the auditory pathways to area 41, which is the primary auditory cortex (Heschls gyrus). From there, they continue to Wernickes area, where the meaning of the words is extracted. In order to speak, the meanings of words are sent from Wernickes area via the arcuate fasciculus to Brocas area, where morphemes are assembled. The model proposes that Brocas area holds a representation for articulating words. Instructions for speech are sent from Brocas area to the facial area of the motor cortex, and from there instructions are sent to facial motor neurons in the brainstem, which relay movement orders to facial muscles. In order to read, information concerning the written text is sent from visual areas 17, 18, and 19 to the angular gyrus (area 39) and from there to Wernickes area, for silent reading or, together with Brocas area, for reading out loud. This model is now obsolete. Nevertheless it has been very useful in directing research and organizing research results, because it is based on the idea that language consists of two basic functions: comprehension, which is a sensory/perceptual function, and speeking, which is a motor function. However, the neural organization of language is more complex than the WernickeGeschwind model of language suggests. The localization of speech in Brocas area is one of the weakest points of this model.[1]

Il segno di ogni lingua dei segni pu essere scomposto in 4 componenti manuali essenziali: movimento, orientamento, configurazione, luogo e componenti non manuali: espressione facciale, postura e componenti orali (non hanno nulla a che vedere con i fonemi del linguaggio parlato).
FoxP2
FoxP2 espresso in particolare nel talamo, gangli della base e cervelletto (studi nel topo). La sua espressione aumenta in alcune aree dei gangli della base in uccelli canori, durante l apprendimento del canto. Ci suggerisce che sia un gene importante per la plasticit dei circuiti che mediano l apprendimento di sequenze motorie speciespecifiche.
FoxP2 expression is increased during times of vocal plasticity. Zebra finches learn to sing by imitating the song of an adult tutor. During the sensitive learning phase birds memorize the tutor song but vocalize little. During the sensorymotor phase they start singing and use auditory feedback to modify their imperfect rendition of the memorized tutor song. This process culminates in a final, crystallized song. (a) In zebra finches, adult song changes little, in contrast to canaries who continue to modify their song throughout life. (b) Before and after the breeding season they incorporate new syllables into their song, which correlates with seasonal plasticity in the neural circuits that mediate the learning and/or production of song. (c) The anatomy and connectivity of the song circuit. HVC and RA are part of the motor pathway necessary for song production (blue). HVC also provides input to the anterior forebrain pathway (AFP) (red). The AFP comprises the nuclei Area X, LMAN and DLM. It is essential for song learning during development and for periods of song plasticity in adulthood. FoxP2 expression in Area X is elevated during times of song plasticity both in juvenile zebra finches (b) and in adult canaries (c).
