UTILIZZO
Il kit fornisce i reagenti necessari per la trascrizione inversa e lamplificazione della regione 5-UTR del virus dellEpatite C (HCV RNA) attraverso una singola reazione. Gli amplificati ottenuti possono essere utilizzati per determinare il genotipo di HCV (codice NLM AC004). Questo prodotto va utilizzato in abbinamento al kit di estrazione di RNA (codice NLM AA077/C) Qiamp Viral RNA Mini Kit Qiagen. Il kit da utilizzare in abbinamento allenzima di amplificazione con tappo di colore verde.

MODULARITA'
La modularit prevista di 4 sedute da 5 test ciascuno. Se la modularit non viene rispettata i reagenti potrebbero essere insufficienti per effettuare il numero di test previsto. Il kit da utilizzare in abbinamento allenzima di amplificazione con tappo di colore verde.
STABILITA E CONSERVAZIONE
I reagenti sono stabili fino alla data di scadenza indicata sull'etichetta se conservati a -20C. Mantenere i reagenti e campioni ad RNA in ghiaccio mentre si prepara la mix.

PRECAUZIONI
La procedura va eseguita utilizzando le buone pratiche di laboratorio ed i comuni dispositivi di protezione individuale Tutti i consumabili (puntali e provette) devono essere sterili. I puntali devono avere il filtro per evitare la contaminazione delle pipette. Utilizzare un nuovo puntale ogni volta che viene dispensato un volume Eliminare il materiale monouso utilizzato, i guanti indossati e tutti i reattivi come rifiuti speciali. Non mangiare, bere, fumare o applicare cosmetici nelle aree preposte allesecuzione del test. Non utilizzare reagenti scaduti. Non mischiare reagenti di lotti diversi. Riportare alla temperatura di conservazione indicata tutti i reagenti subito dopo il loro utilizzo. Prestare attenzione alla modularit del dispositivo (5 test per 4 sedute). Lutilizzo del kit con una modularit inferiore determina linsufficienza dei reagenti per poter eseguire tutti i test previsti. Si consiglia di eseguire lanalisi in tre zone separate: o Zona 1: pre-PCR (manipolazione dei campioni ed estrazione) o Zona 2: preparazione della mix o Zona 3: post-PCR (PCR e rivelazione) E opportuno assicurare una temperatura il pi possibile costante ed uniforme in laboratorio ed evitare di posizionare gli strumenti in prossimit di fonti di calore/raffreddamento che possano comprometterne il corretto funzionamento.

Preparare la mix per il numero di campioni estratti + 1. Miscelare delicatamente e dispensare 35 l di mix nelle provette da 0,2 ml precedentemente contrassegnate. Aggiungere in ciascuna provetta 25 l del rispettivo RNA estratto e mescolare accuratamente. Mettere le provette nel termociclatore dopo avere impostato il seguente programma:

Conservare gli amplificati a 4C se il test di genotipizzazione HCV viene effettuato lo stesso giorno, altrimenti conservare gli amplificati a -20C.
RIVELAZIONE SU GEL (I reagenti per la preparazione del gel non sono inclusi)
Questa fase non richiesta, ma prima di effettuare il test di genotipizzazione HCV possibile rivelare la presenza degli amplificati caricando il prodotto di PCR su gel di agarosio al 2%. I campioni HCV positivi presentano una banda di 233 bp.
TROUBLESHOOTING
In caso di contaminazione importante scoprirne lorigine: estrazione, PCR e/o contaminazioni derivanti da altri amplificati. Lutilizzo di controlli negativi e positivi di HCV possono essere utili nel trovare la causa di contaminazione.

PURPOSE
This kit provides reagents to perform reverse transcription and amplification of 5-UTR region of Hepatitis C virus RNA in one single step reaction. The amplicons obtained can be used to determine HCV genotype (NLM code AC004). HCV RNA must be isolated by using extraction kit Qiamp Viral RNA Mini kit, Qiagen (NLM code AA077/C). Amplification enzyme assigned: green cap.

PRECAUTIONS
Handle this product according to established good laboratory practices and universal precautions; wear personal protective apparel. All disposable items (tips and tubes) must be sterile. Use aerosol-resistant pipet tips to avoid pipettes contamination. Use a new tip every time a volume is dispensed. Discard all used material as biohazardous waste. Do not use components beyond the expiration date indicated on the kit box Do not mix reagents from different lots. Return enzymes to the appropriate storage condition after preparing the working reagents. Pay attention to device modularity (5 tests for 4 assays): if the modularity is not respected, the reagents will be not enough to process all the tests. Do not eat, drink, smoke or apply cosmetics in areas where reagents of specimens are handled. Make analysis in three different areas: o Area 1: pre-PCR (samples handling and extraction) o Area 2: Mix preparation o Area 3: post-PCR (amplification and genotype determination)

It is advisable to have constant and uniform laboratory temperature, avoid to place the instruments near heating/cooling sources that may compromise the right working.

Prepare the mix for the number of the samples to be analysed +1. Dispense 35 l of this mix in each PCR tube previously marked. Add to each tube 25 l of extracted RNA and mix carefully. Put the tubes in the thermal cycler after setting the following program: Cycle 37C 94C 94C 50C 72C 4C Time 45 min 5 min 30 sec 30 sec 45 sec

Keep amplicons at +4C if HCV genotyping is carried out the same day, otherwise store them at -20C. GEL DETECTION (Reagents for gel preparation are not included) This step is not required, but before to proceed with genotyping it is possible to detect the presence of HCV amplicon by loading PCR product on 2% agarose gel stained with Ethidium Bromide. HCV positive samples have a 233 bp band.
TROUBLESHOOTING
When contamination events occur it is important to discover their origin: extraction, PCR reactions or contamination derived from other amplicons. The use of HCV positive and negative controls can be useful to find out the cause of contamination.
