1. BIOMIX (BA071)
La Biomix prevede laggiunta di biotina ad amplificati provenienti da metodiche non-biotinilate. Prima della fase di denaturazione dispensare 4l di Biomix ai 30l di ogni amplificato; miscelare con cura, aggiungere 30l di denaturante, incubare 5 minuti e procedere come da metodica. Attenzione la Biomix viene fornita con una modularit di 4 sedute da 5 campioni ciascuna. Lesecuzione di un minor numero di test per seduta comporta linsufficienza dei reagenti per i test previsti.
2. INNER-MIX (BA064)
La Inner-mix permette la ri-amplificazione con primers biotinilati della regione 5-UTR del virus dellEpatite C, a partire da cDNA o da amplificati ottenuti attraverso altre metodiche di amplificazione virale (metodo nested). La mix da utilizzare in abbinamento allenzima di amplificazione dal tappo color verde.
PROCEDURA
Gli amplificati ad alto titolo (fino a 10 ) vanno diluiti 1:1000 con acqua distillata mentre per quelli a titolo 4 3 2 inferiore (10 ) sufficiente una diluizione 1:100; i campioni a titolo molto basso (10 o 10 ) vanno diluiti 1:50 o 1:10. Data la variabilit di efficienza della precedente amplificazione, tali indicazioni sono prettamente generiche e devono essere adattate a cura dellutilizzatore. Attenzione: per evitare contaminazioni, eseguire le diluizioni degli amplificati e aggiungerli alla mix solo nella zona post-PCR.

Preparare la Master Mix per il numero di campioni +1 volume (per avere una quantit di mix sufficiente per tutti i campioni da processare). Miscelare e dispensare 40 l di Master Mix in provette da 0,2 ml precedentemente contrassegnate. Aggiungere in ciascuna provetta 10 l del rispettivo amplificato opportunamente diluito e miscelare pipettando su e gi. Preriscaldare il coperchio del termociclatore prima di inserire le provette; posizionare le provette nel termociclatore ed impostare il seguente profilo termico:
RIVELAZIONE SU GEL (i reagenti per la preparazione del gel non sono inclusi) Questa fase non richiesta, ma prima di effettuare il test HCV Genotyping possibile rivelare la presenza di amplificati di HCV caricando il prodotto di PCR su gel dagarosio al 2%. I campioni positivi di HCV amplificati con la Inner-mix danno una banda di 238 bp.

The Biomix allows biotinilation of amplificated products coming from non-biotinilated tests. Before starting the denaturation step, add 4l of Biomix in 30l of each amplificated sample; mix well, add 30l of denaturing solution and proceed as previously described. Reagents are provided for four distinct runs of 5 samples each. If modularity isnt respected reagents could not be enough for the number of tests declared
2. INNER-MIX (BA064)
The Inner-mix allows re-amplification with biotinilated primers of 5-UTR region of Hepatitis C virus from cDNA or amplification products derived from other viral amplification methods (nested PCR). The amplicons obtained can then be used to determine HCV genotype through reverse hybridization. Amplification enzyme assigned: green cap
PROCEDURE
High titer amplicons (up to 10 ) have to be diluted 1:1000 with distilled water, whereas those with a lower titer 4 3 2 (10 ) have to be diluted 1:100; samples with a very low titer (10 or 10 ) have to be diluted 1:50 or 1:10. Due to the variability in efficiency of the previous amplification, these indications are to be considered as general and have to be adapted by the single user. Warning: to avoid cross-contamination dilute amplicons and add them to the Inner master mix only in the post-PCR area. Prepare the Master mix for the number of samples to be analyzed +1. Dispense 40 l of this mix in each PCR tube previously marked. Add 10 l of the sample (properly diluted) to each tube. Pre-warm the thermal cycler lid and put the tubes in the thermal cycler after setting the following program:
GEL DETECTION (Reagents for gel preparation are not included) This step is not required, but before proceeding with HCV Genotyping it is possible to detect the presence of HCV amplicon by loading PCR product on 2% agarose gel stained with Ethidium Bromide. HCV positive samples amplified with the Inner mix give a 238 bp band.
STRIP ASSAY Detection as described in AC004 with 15l of amplificated product and 15 l of DNAT.
