Nei prodotti a shelf life brevissima talvolta non si fa nemmeno il trattamento termico, perchè ho stimato che in
quella breve shelf life non possa crescere nulla di pericoloso.
Elementi di giudizio:
In generale (indipendentemente dalla tipologia di materia prima):
- Caratteristiche del prodotto finito (pH, aw, inibitori chimici)
- Caratteristiche del contenitore (permeabilità all’O2, rilascio di nutrienti)
- Tipo di shelf-life (temperature e tempi di mantenimento)
Esclusivamente per alterativi molto termoresistenti (in genere termofili obbligati):
- Materie prime impiegate e loro origine
- Fasi di lavorazione pre-sterilizzazione (aumento/diminuzione della carica sporale)
- Temperatura ambiente massima di conservazione e distribuzione prevista
- Destinatari del prodotto (figure professionali, tipo di consumatore)

Criteri per la distruzione microbica
"Low-acid food" means any food, other than alcoholic beverages, where any component
has a pH value greater than 4.6 and a water activity greater than 0.85.
"Acid food" means a food that has a natural pH of 4.6 or below.non abbiamo problemi con il Botulino

"Equilibrium pH" is the pH of the macerated heat processed food product.

"Commercial sterility of thermally processed food" means the condition achieved by
application of heat, sufficient, alone or in combination with other appropriate treatments, to
render the food free from microorganisms capable of growing in the food at normal non-
refrigerated conditions at which the food is likely to be held during distribution and storage.

Bacterial food-borne pathogens may be grouped into those that cause food
intoxication and those that can result in food-borne bacterial infection.
In case of bacterial food poisoning or intoxication the causative organism multiplies in the
food where it produces its toxins.
Thus ingestion of viable bacteria is not a prerequisite for the induction of the disease.
Most often intoxications require that the toxin producing bacteria have grown to high
numbers (105 – 108 cfu/g) in the food before it is eaten.
In contrast, the food merely act as a carrier for the causative organism in food-borne
infections.
The infectious agent may or may not have multiplied in the food, but the ingested viable
bacteria continue to grow within the host’s body to produce the typical symptoms (fever,
diarrhoea).
The number of viable bacterial cells necessary to cause disease (the Minimum Infective
Dose, MID) varies considerably between bacterial species.
The current Good Manufacturing Practice Regulations (GMP's) for low-acid canned
foods (21 CFR 113) and the GMP's for acidified foods (21 CFR 114) outline the
equipment, controls, manufacturing, processing, and packing procedures which are
required to ensure the production of a safe product.
The purpose of the above regulations is to protect consumers from microorganisms of
public health significance (i.e., harmful bacteria or their toxins), especially Clostridium
botulinum. The spores of Clostridium botulinum must be destroyed or effectively
inhibited to avoid germination and subsequent production of the deadly toxin which
causes botulism. This is accomplished with good manufacturing practices which must
include:
The use of thermal processes and/or other means of preservation that are
properly designed by a competent processing authority to destroy or inhibit
Clostridium botulinum spores;
Proper delivery of these thermal processes and/or adequate control of other
methods of preservation being used.
Sufficient documentation that the scheduled processes are delivered through the

LOW-ACID FOOD: Any food (other than alcoholic beverages) with a finished equilibrium

vary, for example, by 1 C could result in a difference in aw reading of 0.05.
The water activity level of 0.85 is used as a point of definition for determining whether a
low-acid canned food or an acidified food is covered by the regulations. Low-acid
canned foods can be preserved by controlling water activity at levels above 0.85.
The minimum aw level for the growth of C. botulinum is approximately 0.93. Depending
on various product characteristics this minimum level can be as high as 0.96.
The regulations (21 CFR 113.3(e) (1) (ii)) state that commercial sterility can be achieved
by the control of water activity and the application of heat.
The heat is generally necessary at aw levels above 0.85 to destroy vegetative cells of
microorganisms of public health significance (e.g., staphylococci) and spoilage
microorganisms which can grow in a reduced aw environment.
(f) The scheduled thermal processes for foods having an aw greater than 0.85 and less
than the aw that would allow the growth of spores of microorganisms of public health
significance (**) shall be sufficient to render the food free of microorganisms capable of
reproducing in the food under normal nonrefrigerated conditions of storage and
distribution.
Growth can be influenced depending on the composition of the product. The main

Preservative agents: curing agents for meat products, grow limiting additives such as
lactate, etc. Combinations of pH, aw and curing (preservative) agents.
Mesophilic strains of B. cereus can grow between 10 ºC and 42 ºC, some strains being
able to grow at 50 - 55ºC.
Phychrotrophic strains would grow at temperatures as low as 4 ºC, with optimal growth
temperatures between 30 and 37 ºC and maximum ranged from between 37 and 42°C.
Refrigeration reduces growth of B. cereus by increasing the generation time (1.6 h at
19.5°C, 2.9 h at 14.2°C, 4 h at 9.6°C and 6.7 h at 6.5°C). Refrigeration also considerably
increase lag time (148.77 h and 1.96 h at 5 and 30 ºC).
pH limit for growth in carrot substrate acidified with citric acid was between 4.5 and 4.75.
In milk acidified with HCl, slight growth was observed at pH 4.1 at 37°C. At 25°C, growth
rate dropped dramatically below pH 5.5.
The water activity must be higher than 0.92 for growth.
In modified atmosphere packaging growth rate was reduced as the proportion of CO2 in
the gas mixture increased.
Nisin produces the inactivation of Bacillus cereus vegetative cells.
Spores of B. cereus have a broad range of heat resistance (D100 = 1,8 - 3,9). Acidification
from pH 7 to 4 produced a five fold decrease in D103ºC values.

Use of ROP (Reduced Oxygen Packaging) with some foods can markedly increase
safety concerns. Unless potentially hazardous foods (time/temperature control for safety
foods) are protected inherently, simply placing them in ROP without regard to microbial
growth will increase the risk of foodborne illnesses. (FDA Food Code 2009 – Amnnex 6)

The principle of the method specified is preparing the test sample according to the class to
which the product to be examined belongs, and measuring the potential difference between
a glass electrode and a reference electrode immersed in the test sample (a 25°C).
Applies to four classes of product:
1. homogeneous products with a liquid or thick texture, or products exhibiting a large
liquid or thick phase; (mescolare prima della misura)
2. homogeneous pastes or heterogeneous products for which homogenization is

Performance Standards for the Production of Processed Meat and Poultry
Products (Federal Register: February 27, 2001 (Volume 66, Number 39) Proposed Rules)
The 12-D concept arose from studies on the thermal resistance of C. botulinum
conducted in the early 1920's by scientists of the National Canners Association
(predecessor of the National Food Processors Association).
These scientists inoculated a phosphate buffer with spores of the most heat-resistant
strain of the organism then known. They determined, by extrapolating from the
exponential survival curve for the organism, the temperature and duration of the heat
process necessary to reduce the population from 6 x 1011 spore/unit to less than one
spore/unit.
Subsequent studies on products inoculated with C. botulinum and other organisms
essentially confirmed the results of these studies.
These products undergo a botulinal cook to achieve an acceptable safety level.
It should be noted that the intensity of the process is not related to the actual number of
C. botulinum organisms that may be in the product. That number is usually very low in a
meat product (less than a spore per kilogram). So the 12-D process provides a
tremendous safety margin to consumers.
A series of beginning contamination levels for raw food coming into typical foodservice
systems is shown in Table 4-3. It is based on threshold levels that make healthy people ill
and normal contamination as listed in the literature. These figures pertain to food in the
U.S. When designing a safe food process, these are the levels of contamination that must
be controlled and eliminated by methods of processing, preservation, and storage.

good food quality?" Neither the USDA nor the FDA provides an answer to this question.

Using the highest known D-values (0.25 min at 121 °C) the Fo is therefore equal to 12 x
0.25 = 3. This is the so-called 12 D-concept designed to reduce the bacterial load of one
billion spores in each of 1000 cans to 1 spore in a thousands cans.

Survival of Bacillus cereus spores at 95 ºC decreased by three fold when the pH of the
heating substrate was decreased from 6.2 to 4.7 (Fernandez et al 2002). Mazas et al (1998)
found that acidification from pH 7 to 4 produced a five fold decrease in D103ºC values.
B. cereus is ubiquitous and its spores are present in almost all categories of foods.
Dry foods such as dry soups, dried dairy products, infant formulae, spices, herbs and
seasonings are frequently contaminated with B. cereus and other Bacillus species
although at different levels. Rice and pasta dishes have frequently caused emetic
poisoning.
Sterilization is the most effective way to control Bacillus cereus spores.
Considering heat resistance data (Fernandez et al 1999), 3 min at the constant
temperature of 105 ºC can produce 5 log reductions in the population of a high resistant
Bacillus cereus strain. Temperatures higher than 105 ºC should protect food from this
microorganism in most instances. However, only canning can ensure complete
destruction of B. cereus spores.

Essentially, all low-acid foods are processed far beyond the minimum botulinum cook in order to
deal with spoilagecausing bacteria of much greater heat resistance. For these
organisms, acceptable levels of spoilage probability are usually dictated by economic
considerations. Most food companies accept a spoilage probability of 10-5 from mesophilic
spore-formers (organisms that grow and spoil food at room temperature). The organism most
frequently used to characterize this classification of food spoilage is a strain
of Clostridium sporogenese known as PA 3679 with a maximum D250 value of 1.00. Thus, a
minimum process sterilizing value for a mesophilic spoilage cook assuming an
initial spore load of one spore per can is F = 1.00 X 5 = 5.00 min.
Where thermophilic spoilage is a problem, more severe processes may be necessary because
of the high heat resistance of thermophilic spores. Fortunately, most thermophiles do not grow
readily at room temperature; they require incubation at unusually high storage temperatures
(43.3 – 54.4°C) in order to cause food spoilage. Generally,
foods that show no more than 1% spoilage (spoilage probability of 10-2) upon incubation after
processing will show less than the minimum 10-5 spoilage probability in normal
commerce. Therefore, when thermophilic spoilage is a concern, the target value for the final
number of survivors is usually taken as 10-2, and the initial spore load needs to be determined
through microbiological analysis since contamination from these organisms varies greatly. For a
situation with an initial thermophilic spore load of 100 spores per can and an average D250 value
of 4.00, the process sterilizing value required would be
F = 4.00 (log 100 - log 0.01), F = 4.00 (4) = 16 min.
Thermophilic spoilage
It is generally found acceptable if thermophilic spore levels are reduced to around
10-2 to 10-3/g.
There are two reasons why higher risks of spoilage (arising through survival,
germination and outgrowth of thermophilic spores) can he tolerated.
First, given reasonable storage temperatures (i.e., <35 °C) the survivors will not
germinate; and secondly even if spoilage does arise it will not endanger public health.
Pflug (*) recommended the following target end points for the lethal treatment:
• Pubblic healt end point of 10-9 for C. botulinum
• Mesophilic spoilage end point of 10-6
• Thermophilic spoilage end point of 10-2 for low temperature storage or 10-6 for high
temperature storage

In funzione del tipo di processo applicato, possono risultare critici per il trattamento
termico del prodotto i parametri relativi a:
- Formulazione (qualitativa e quantitativa)
- Pre-trattamenti
- Condizioni iniziali
- Condizioni del trattamento termico
Per ciascun parametro critico, individuare la variabilità e la condizione che risulta meno
favorevole allo scambio termico.
N.B. Tutti i parametri critici devono essere sotto controllo statistico e la loro variabilità
naturale dipende dalla precisione e dalla sensibilità degli strumenti di controllo impiegati.
Qualora i parametri critici non siano già sotto controllo statistico, è necessario analizzare
i risultati di una prima campagna di acquisizione dati, individuare ed eliminare con adatte
istruzioni di lavoro le cause di deviazione accidentale (cause assegnate) e ripetere la
raccolta dati per individuare la variabilità “naturale” (ovvero quella intrinseca alle
condizioni di lavorazioni esistenti).

The calculator converts temperature readings to lethal rates, plots the lethal rates against
time, and determines F values for a heat process. The area under the curve is determined
using the trapezoid rule. Accurate F determinations for most thermal processes can be
obtained. In general the more values, the more accurate the value for F will be.
The lethal effect of high temperatures on microorganisms is dependent on both temperature
and holding time.
Because microorganisms in foods are exposed to lethal temperatures as they reach the target
processing temperature and during cooling, it is necessary to calculate the cumulative effect of
heat on microbial destruction during both heating and cooling as well at the holding time at the
target temperature.
The lethal rate is a dimensionless number and can be calculated using equation 1 (Stobo,
1973). Lethal rate = 10(T-Tr)/z where T is the temperature, in Celsius, at which the lethal rate is
calculated and Tr is the reference temperature at which the equivalent lethal effect is
compared. The z-value measured in °C is the reciprocal of the slope of the thermal death
curve for the target microorganism or spore; 10°C is the value frequently used in Fo
calculations performed on low acid foods. Note the original work on themobacteriology used a
reference temperature of 250°F. This is equivalent to 121.11°C. Tr will vary depending on
whether Fo is being calculated or whether a pasteurisation process or other heat treatment is
being assessed. A Tr value of 121.11° C is used in the determination of Fo. If F70 or other F
value is required then Tr can be set at 70°C or other temperature.

Per poter essere applicato ai fini commerciali, il processo deve essere formalizzato in
procedure e istruzioni di lavoro in maniera tale da garantirne continuativamente la
corretta applicazione.
La procedurizzazione, che deve avere una codificazione univoca rispetto al prodotto
ed alle sue eventuali varianti di formulazione e/o di linea di processo, deve
comprendere l’indicazione di tutti i parametri critici, con i corrispondenti valori fissati,
le modalità di tenuta sotto controllo e quelle di verifica.
Obiettivo:
Garantire la sicurezza igienica e la stabilità microbiologica con:
• la migliore qualità nutrizionale e sensoriale
• i minori costi di produzione (meno energia ÷ più capacità produttiva)
Criteri metodologici:
1. Ridurre la variabilità degli effetti inibitori che incidono direttamente sulla qualità

Nel caso della aw, che in genere è condizionata sia dal contenuto di acqua sia da quello

Nei derivati del pomodoro, le spore di C. pasteurianum (il ceppo di butirrici più
termoresistente) a pH = 4,3 hanno: D90 = 7,19 minuti e z = 9,5°C, quindi D100 = 0,64.
A pH = 4,3 devono sopravvivere più di 102 spore per confezione per avere la probabilità
che si alteri più di una confezione su 104.
Utilizzando come materia prima pomodoro in condizioni igieniche normalmente buone
(assenza di terriccio alla triturazione), il valore di carica iniziale può essere di 10-2 spore
butirriche per grammo; ovvero circa 105 spore in un sacco asettico da 200 kg.
Per ottenere un prodotto microbiologicamente stabile a pH = 4,3 (con non più di 102 spore
per confezione), occorre applicare con il trattamento di pastorizzazione 3 riduzioni

In any finite mass of liquid (or any material), the temperature history T(t) varies spatially
throughout the mass. Thus F0 will vary spatially at any given time. Thus in reality, there is
always a distribution of F0 values due to spatial variation of temperature history.
For bacterial destruction the lowest value of F0 is taken as the measure of the extent of
sterilization. The average F0 for nutrients, which is sometimes referred to as the cook
value, provides a measure of the nutritional quality of the processed food material.
The complete distribution of quality parameters such as sterilization and nutrient retention
is generally quite difficult to obtain due to the complex variations of temperature T with
position and time in most processing situations of practical interest. Under these
circumstances, design of the heating process is aimed at minimizing the spatial variation
of temperature, for example, by introducing turbulence.
