Maintenance of syringe mechanisme

bbbb 13
Maintenance of optical mechanism

bbbb 14
Maintenance of optical mechanism
• Lamp adjustment
• Photo-sensor adjustment
• Cuvette-rotor adjustment
• Cleaning the optical filters
• Replace optical filters when
needed

bbbb 15
Reagent needle ball bearing
• When got rusty, replace the bearing
• Bearing gets rusty when it often gets wet, probably caused by bad
adjusted PLL.

bbbb 16
Vacuum pump membranes
• Membranes should be replaced every half year
• There are two types available
– Gray membrane pn: 6002-040
– Red Membrane pn: 3913-049
• Chamber kit should be replaced every two years
• There will almost always be a little liquid in the system, due to the
design of the system (waste gasks)

bbbb 17
2-way valves
• Consumption two-way valve is about 100/year/5000instr
• Consumption three-way valve is about 100/year/5000instr

• Relatively higher consumption of two way valves. Probably due to

the fact that the two-way valve is more in contact with chemistry
waste then the other valves.

• Relative consumption is less then one valve per instrument per 10

year

bbbb 18
Syringe tips
• Regular lifetime of a sample tip should be 4-6 weeks (depending on
the workload)

• Vital is running experiments at the moment with an other supplier of

syringes

• Replacement of the glass-barrel or the complete syringe is part of

the half yearly maintenance. And strongly recommended

bbbb 19
Air bubbles in the water system
• Air bubbles are trapped in the tubing system
• Use of fresh tapped de-ionized water contains a lot of dissolved
water
– Use a spare canister of water, so dissolved air can escape from
the water
• Use of system liquid avoids air bubbles sticking to the tubing

bbbb 20
Maintenance kits suggestions for
• ½ Yearly kit
improvement
• Yearly kit
• Food analysis instruments

bbbb 21
SD_error on reaction rotor
• Important not to make scratches
on the rotor when searching for
the slit to fit.
• Important not to touch the
cuvette with the fingers
• In future software it will be
possible to exclude cuvettes from
measurement

bbbb 22
Overfilling the drain positions of the needle
• September 2003 a drain modification has been send out to all XL
instruments (distributors) at that time. The content of the drain was
enlarged

• At the moment it only can be cause by not sufficient draining of the

drain. (obstruction or low vacuum)

bbbb 23
User items
Problems with TP, Mg, Ca, Glucose
• What are the problems
• Collect as much information as possible

• Is there a common item (filter, sample volume)

• What is the SR3 and SR75 performance
• Related to the instrument?
• Related to the reagent?

bbbb 24
User items
How to improve low throughput
• make an inventory of test volume per analyte
• Separate 50% left and 50% right
• Separate incompatible tests, left and right
• Avoid a lot of incompatibilities

bbbb 25
User items
Incompatibilities (which are necessary or
not)
• Incompatibilities are depending on the reagents
• The reagent manufacturer should be able to advise

Lactate Test
LDH
L-Lactate + NAD+ ------------------- Pyruvate + NADH + H+

Measured is the increase of NADH at 340 nm

LDH is part of the reagents and present in a high concentration
This LDH of the reagent could falsely elevate a patient LDH test

bbbb 26
User items
Incompatibilities (which are necessary or
not)
ALT Test
ALT
L-alanine + α-ketoglutarate ---------------------- L-glutamate + pyruvate
LDH
Pyruvate + NADH + H+ --------------------------- Lactate + NAD+

Underlined items are part of the reagent and in a high concentration

Measured is the decrease of NADH at 340nm
LDH is in a high concentration present in the reagent contamination
The LDH of the reagent could give a falsely elevated result of the LDH test

bbbb 27
User items
Incompatibilities (which are necessary or
not)
TRIG Test
Lipase
Triglicerides --------------------------- Glyserol + Fatty Acids
GDH
Glycerol + NAD+ -------------------- Dihydroxyacetone + NADH + H+

Underlined items are part of the reagents

In some reagents from other tests there is glycerol present as a kind of
preservative. This can give in a contamination falsely elevated results of
the TRIG test itself
(BUN, AMY URCA)

bbbb 28
User items
Selective unload possible?
• NO when bar-code option is switched off

bbbb 29
User items
Printer compatibility problems
• Flexor E and XL use an external printer. When the correct driver is
installed there should be no problem
• Only one complete patient report per page
• When report set-up is used, a non-proportional letter-type should be
use (Courier New, San Serif)
• Pictures factor horizontal 6 and vertical 6

bbbb 30
User items
Printout sequence
• Printer will print in the order of measurement and only completed
patient reports.
– This can be disturbed by a remeasurement
– To print e.g. no 15 before 1-14 is only possible by requesting
manually a print by F1(in evaluate samples)

• Future software will be capable to give a re-measurement a

higher priority

• Future software will be capable to print more then one patient

report per page.

bbbb 31
User items
Sample and dead volume
• Dead volume on cup 50ul
• Dead volume on tubes with standard rotor 500ul
• Low dead-volume rotor (E) pn: 6002-121 200ul

• Sample volume to high?

– Depends on reagents
– Depends on total cuvet volume

bbbb 32
User items
Programming applications
• Read insert sheet
• The used wavelength should be mentioned
• Should indicate endpoint, two point or kinetic
• Should give pipetting volumes
– Reduce these volume to min-max cuvette volume
• If test is a long endpoint, maybe it could run as a kinetic test
– This can be tested to run it as kinetic and use the longest reading
possible

bbbb 33
User items
When are the reagents, controls and standards
OK?

bbbb 34
User items
When are the reagents, controls and
standards OK?
• Calibration on 1 standard in duplicate
• Calibration on 1 standard also a reagent blank
• When a drift is seen on the QC graphic
– Recalibrate
– Refresh reagents

bbbb 35
User items
Power schemes of the PC and Monitor
• Vital advises to keep them switched on
– Screen can be switched off by its main-switch

• When switching them off

– Switch off the cooling unit (condense)

bbbb 36
User items
ISE module disadvantages and
incompatibilities
• Minimum of samples / day = 25
• Ratio human sample : calibrators/controls
20 : 1
• Model 9130 VS is only compatible with Flexor / Selectra
• Q1 2005 Dry ISE will be introduced
