Connective tissue cells

First type:
Fixed or permanent cells
Are native to the tissue in which they are found:

Undifferentiated mesenchymal cells


Fibroblasts
Fibrocytes
Macrophages
Fat cells
1- Undifferentiated mesenchymal cells:

- Cytoplasm is faint basophilic.


- Theyre small stellate cells with thin cytoplasmic processes.

- Function: Theyre STEM CELLS, can divide and differentiate into


many types of CT cells.
2- Fibroblasts

- Most common cells in Ct.


- Are spindle- shaped branching cells.
- Basophilic cytoplasm, rich in rER & well developed Golgi complex.
- Have large euchromatic, oval nucleus
- Function: synthesis and secretion of CT Matrix.
3- Fibrocytes

- Have small size, dark nucleus with fewer rER.


- Once fibroblasts synthesize in the matrix, they become less active
fibrocytes.

- Function: maintenance of connective tissue matrix.


4- Macrophages

Are large irregular with pseudopodia.


Are derived from the circulating blood monocytes which leave the blood
stream to migrate into the CT.
Have kidney-shaped eccentric nucleus and eosinophilic cytoplasm with
many lysosomes and phagosomes.

Function: phagocytosis of bacteria, cell debris, malignant and virus


infected cells.
5- Fat cells

Are large cells, polyhedral in shape.


By H&E, The cytoplasm is empty ( The large single lipid droplet is dissolved
during histological slide preparation ) with peripheral flattened nucleus
signet ring appearance.
Stain : Osmium tetroxide, adipocytes appear black in color.

Function: Storage of fat.


Second type:
Wandering Cells
Are immigrant cells usually from blood or bone marrow.
Some retain their own characteristics and may take up
permanent residence there:

Blood leukocytes
Mast cells
Plasma cells
6- Leukocytes

Theyre circulating in blood stream and then migrate into the CT to perform
their specific function.

Types: neutrophils, eosinophils, basophils, lymphocytes and monocytes .


7- Mast cells

The cytoplasm is full of basophilic granules that may obscure the central
nucleus.
A staining feature is Metachromasia i.e. by toluidine blue gives a purple color
instead of blue, due to the acidic glycosaminoglycans of their secretory
granules.

Function:
These cells can initiate local inflammatory response when release the content of
their secretory granules. These includes:
1- Heparin : as anticoagulant
2- Histamine : causes allergic reaction (edema, hypotension, bronchospasm and
skin rash)
