Protein Pharmaceuticals

-Protein pharmaceuticals is a manipulated form of


protein, which is generated in various ways to produce
large quantities of proteins, modify gene sequences and
manufacture useful commercial products.
-Protein engineering is the process of developing useful
or valuable proteins. It is a young discipline, with much
research taking place into the understanding of protein
folding and recognition for protein design principles.
--Enzyme engineering is the application of modifying
an enzyme's structure (and, thus, its function) or
modifying the catalytic activity of isolated enzymes to
produce new metabolites, to allow new (catalyzed)
pathways for reactions to occur, or to convert from some
certain compounds into others (biotransformation).
Types of Protein Pharmaceuticals

a. Protein Pharmaceuticals from blood-Our body is


composed of 60%-70% plasma and 8%-9% is protein.
Plasma contains many different proteins.

Here are some of the examples:


Factor VIII (For blood-clotting for hemphilia)
Factor IX (For blood-clotting for hemophilia
Albumin (osmotic balance-for kidney disease)
Ig IV (For treating infections)
Anti-Thrombin III (For blood-clotting)
Alpha I-PI (For Emphysema)
Types of Protein Pharmaceuticals

b. Monoclonal antibodies-This are very specific for


certain antigens and cell types and can be used to target
foreign toxin, viruses or cancer cells. They are used for
drug delivery to specific targets and have a half-life often
greater than one week when humanized.

c. Peptide drugs-Many hormones are actually small


peptides (2-40 amino acids) and as such, peptides can be
synthesized and can be used as pharmaceuticals.

-The formation of recombinant protein is carried out in


specialized vehicles known as vectors. Recombinant
technology is the process involved in the formation of
recombinant protein.
Two ways of generating Protein Biopharmaceuticals:

2. expression in transgenic plants/animals

DNA Biopharmaceuticals

-Vaccines help the body fight infectious diseases.


Traditional vaccines may be live attenuated, where
pathogen is weakened through chemical or physical
process and whole pathogen that is completely killed
or destroyed chemically.
-A new generation of vaccines is the recombinant
DNA vaccines. DNA coding for an antigen is
incorporated into a plasmid and the resulting
recombinant plasmid is administered into the patient.
