Experiment Goal
•

• To observe the work of catalase enzyme in

various substrate and various pH

Early Observation
 Catalase is an enzyme found in nearly every
cells
 Produced by an organell named Peroxixome.

 Convert hydrogen peroxide to water and

oxygen

• H2O2 → H2O + O2
•

Early Hypothesis
 Enzyme Works on Specific pH
• Therefore, catalase enzyme do work in
specific pH as well.
•
∴ Theoptimum pH for catalase
enzyme is 7 (neutral)
•

 Catalase Mostly Found Only in Specific Cells

• catalase enzyme is produced by an
organelle named Peroxixome.
• Peroxixome mostly found in liver cells.
•
∴ Catalase will have bigger
concentration in liver cell 4
Tools and Materials
 Reaction tube (9 pcs)
 Ruler (1 pc)
 Substrate:
 Crushed liver of Gallus gallus (chicken)
 Crushed Carrot
 Crushed Potato
 Hydrochloride Acid (HCL)
 Sodium Hydroxide (NaOH)
 Hydrogen Peroxide (H2O2)
 Plastics (9 pcs)
 Ribs (± 10 cm)

Procedures
 Prepare all materials and tools
 Put crushed liver, crushed potato, and crushed
carrot in each three reaction tube.
 Name the tube for each substrate with A, B,
and N.
 Give 10 drops of HCL in tube A, 10 drops of
NaOH in tube B for each substrate. Let tube
N untouched.
 Give 10 drops of H2O2 in all tubes. Quickly wrap
the tube with plastic and bundle it with rubber
 Wait for about 300 seconds (5 minutes).

Procedures
 After 5 minutes, open the plastic wrap and do
“Gas Test” by using ember ribs for all tubes.
 Pay attention to the ember of the ribs.

Conclusion
 Catalase enzyme work best in neutral
condition. This is proven by the lighter
ember in neutral-substrate during Gas Test.


 Catalase enzyme have higher concentration in

liver cell. This is proven by the lighter ember
in chicken liver than the other substrate.


 Catalase enzyme do react with hydrogen

peroxide forming O2 gas. This is proven by
the lighter ember during gas test.
