TITLE: Molecular characterization of ferritin gene and phytic acid in rice with specific reference to its deposition in endosperm

ABSTRACT
The present investigation is intended to do the molecular characterization of ferritin gene and phytic acid in rice with specific reference to its deposition in endosperm. We have choosen this work because iron deficiency is the most wide spread nutrition disorder in world. The distinct strategies have been developed to improve iron density to increased the grain iron content and its bioavailability so fur. The iron content in seed may be improved through breeding approach, using iron and non iron content varieties. In this investigation, recipient varieties are NDR359, CSR13, SWARNA, SWARNA SUB1, and donor varieties are SARJOO52 and MADHUKAR. These selected varieties will be studied in reference to iron and phytic acid concentrate per standard .protocol. Ferritin gene will be quantified by isolation of total RNA from both the parents and Fl, using RTPCR. Protein and carbohydrate percentage from iron and non iron content varieties will be also correlated with iron deposition in the endosperm. The overall work will be done in Narendra Deva University of Agriculture & Technology, Kumarganj, Faizabad (U.P.).

Introduction:
Rice (Oryza sativa L.) is one of the most important food crop in Asia. Rice is cultivated all most world wide over an area of about 153.51 millions ha. With an annual production about 614.65 millions tones with an average productivity of 4.00 millions tones. Rice is generally used by 50% of the world population in dependent on it for daily carbohydrates. India has largest area (44.6 m ha) constituting 28.0 1% of the land under rice production in the world (FAO, 2008). Iron deficiency, is the most widespread nutritional disorder in the world and India is no exception. More than two billion people globally are iron deficient, and the problem is severe enough to cause anemia in 1.2 billion people (Misra et al, 2004). It has been estimated that 40% of the worlds women suffer some degree of iron deficiency. Ferritin protein is an iron storage protein found in plants, animal and bacteria, which have ferroxidase activity. Phytic acid is major component of cereals grain and beans and is

considered and important antioxidant. Phytic acid is essential for germination of rice but decrease the iron content in rice. Objective of investigation: 1. Qualification for ferritin gene via RT-PCR in rice. 2. Quantification for phytic acid in back crosses line.
3. Searching suitable DNA marker/protein band correlating with adequate ferritin

gene. 4. Correlating with protein & carbohydrate in line containing iron. Technical program:1. Method of approach for first objective: A. Isolation of total RNA. B. cDNA synthesis from DNase treated RNA. C. PCR. Amplified. 2. 3. Method of approach for second objective: A. Quantification of phytic acid as per standard protocol. Method of approach for third objective: A. Isolation of DNA. B. PCR amplified with marker. C. Identification and amplified /novel band & Cloning with desired product. 4. Method of approach for fourth objective: A. Protein isolation for iron and non iron containing variety, f1 and back cross. B. SDS-PAGE analysis of protein for step A. C. Weston blotting of using ferritin antibody.
