Determination of KLa

Objective: To determine KLa value in a fermenter at given impeller rotational speed and gas flow rate using static method of gassing out.

Apparatus required:
25 L aerated stirred fermenter DO2 probe and sensor Temperature probe and sensor pH probe and sensor N2 gas cylinder Rotameter Tachometer stop watch Materials required: Corn Steep Liquor 300 ml DM water 14.7 L

System variables: Temperature 37 oC pH 7.0 Impeller rotational speed 100 rpm Gas flow rate 1 vvm Pressure 1 atm., gauge

Procedure: 1. 15 L of CSL (5% w/w) is poured into the fermenter. System variables are adjusted as per specified values above. The vessel is aerated at 1 vvm and stirred at 100 rpm for 30 minutes. 2. DO2 probe is calibrated. 3. N2is flushed such that Dissolved oxygen reads 0%. 4. Air is sparged into the vessel at 1 vvm and pressure is adjusted to 1 atm., gauge. 5. As soon as all the specified conditions have been restored, DO2 reading is taken for time t=0. 6. After every 15 seconds readings are taken until the dissolved oxygen reaches 80%. 7. If the reading changes slowly, interval of time can be increased accordingly. 8. Graphical analysis is done using Microsoft excel and KLa value is obtained using least square fitting method.
