future.2.13. Identificacién de las bacterias sulfato reductoras por técnicas
moleculares
La identificacion de las BSR sobre la base de sondas y cebadores de DNA o RNA
han sido usada en diferentes trabajos, ya sea mediante la amplificacién del gen
que codifica la enzima sulfito reductasa_desasimilatoria (dsr AB) 0 el gen DNAr
16S (Steuber et al., 1995; Wagner et al., 1998; Minz et al., 1999; Larsen et al.,
2001).
2.13.1. La sulfito reductasa disimilatoria
La sulfito reductasa disimilatoria (DSR) muestra una estructura terciaria
compuesta de dos subunidades proteicas llamadas dsrA y dsrB, organizadas en
dos grupos, formando un tetramero con un “siroheme” en el dominio dsrA y dos
grupos prostéticos Fe-S en ambos dominios (Steuber et al.,1995)
Larsen y colaboradores (2001) observaron que el grupo de genes en
Thermodesulforhabdus norvegica se ordenan en seis marcos de lectura abierta
(ORC's): ds/D-dsrA-ds/B-dsN-dsrC-fdx. La mayor parte de la proteina
ensamblada corresponde a las subunidades dsrA y dsrB, con pesos de 49kD y
43kD respectivamente, mientras que las subunidades dsrD, dsrC y dsrN son
mucho més livianas (aproximadamente entre 13kD) y intervienen en la reduccién
de sulfito a sulfuro (Larsen et al., 2001).
Se estima que el origen de la enzima DSR data de hace aproximadamente 2800
millones de afios y que la divergencia entre dominios ocurrié aproximadamente
hace 3100 millones de ajios. La funcién metabdlica de la DSR en el ciclo global de
azufre es muy importante y se conoce la secuencia completa de las subunidades
A y B de al menos un género de cada divisién microbiana donde esta presente la
reduccion del sulfato (Minz et al., 1999), por lo que fue posible realizar analisis
comparativos del ordenamiento filogenético de las BSR, basados en secuencias
de RNAr 16S y de la DSR (Joulian et al., 2001). Muchos trabajos muestran que
existen suficientes evidencias en el andlisis filogenético comparativo de la DSR y
el RNAr 16S para suponer que la habilidad de reducir sulfato fue transmitida
mediante un proceso de transferencia horizontal antes o durante la divergencia de
los dominios Arquea y Bacteria y posteriormente entre grupos bacterianos gram
positivos y BSR termofilicas (Wagner et al., 1995; Cottrell y Cary, 1999; Pérez-
