Beer's Law and Spectrophotometry:
Beer-Lambert Law gives the relationship between the absorbance of
light in a sample and its concentration. The path length and the molar
absorptivity are held constant (based on the instrument and the
wavelength of light used).

"' '
Beer's Law and Spectrophotometry:
When deciding to do spectrophotometry the first thing one must do is
select the correct wavelength of light to do the experiment at.
=

The correct wavelength is the one that has MAXIMUM

absorbance in a certain material. If you plot absorbance vs
wavelength for a given concentration you get a graph like below.

It's worth noting that colors come from reflecting light back to your
eyes- so something that looks blue will MOST absorb in the orange/
F-

red area of the spectrum and reflect the most blue. This can help you
-

better predict where this should occur.

In a spectrophotometry experiment...
O 1. A calibration curve is created using known concentrations of

El . a particular ion at a fixed wavelength and optical length which

creates a plot of absorbance vs concentration.

2. An unknown with the particular ion is analyzed at the same

wavelength and is compared to the curve. Based on where it

lands on the graph- you can tell its concentration.
