RESUMO
ABSTRACT
The pharmacological induction of ejaculation has been an alternative used to
increase ejaculatory function of stallions incapable of ejaculating by the
traditional methods of semen collection. However, the protocols developed to
date are available in high rate of dose variation, routes of administration,
adverse effects and ejaculation rates. In general, pharmacological protocols
have shown low success rates in inducing ejaculation. Thus, the aims of the
study were: 1) Compare the efficiency of different protocols to induce ex copula
ejaculation when detomidine and oxytocin was added to the protocols; 2)
Compare seminal parameters between in copula and ex copula ejaculates; We
evaluated the nine protocols to ex copula ejaculation: X - xylazine (0.66mg/kg/
i.v); XO - xylazine + oxytocin (20UI/i.v); IX - imipramine (3mg/kg/v.o) + xylazine
(0.66mg/kg/i.v); IXO - imipramine (3mg/kg/v.o) + xylazine (0.66 mg/kg/i.v) +
oxytocin (20UI); D - detomidine (0.02mg/kg/i.v); DO - detomidine (0.02mg/kg/i.v)
+ oxytocin (20UI/i.v); ID - imipramine (3mg/kg/v.o) + detomidine (0.02mg/kg/i.v);
IDO- imipramine (3mg/kg/v.o) + detomidine (0.02mg/kg/i.v) + oxytocin (20 UI/
i.v); IO- imipramine (3mg/kg/v.o) + oxytocin (20 UI/i.v). Four young stallions (2-3
y old) and 12 sexually mature stallions (6 to 26 y old) were each submitted to 2
treatment trials conducted at 3-day intervals. Induced ejaculates were collected
into a plastic bag and compared with in copula ejaculates. None of the 4 young
stallions ejaculated and 9 of the 12 mature stallions responded. Stallions that
responded to xylazine did not respond to detomidine treatments. Two stallions
responded to X and XO while 4 mature stallions responded to IX and IXO. One
stallion respond to DO while 5 stallions responded to IDO. Erection occurred in
5 stallions and no erection and masturbation were observed in protocols without
imipramine. The induced ejaculates obtained in DO and IDO had significantly
(P<0.05) lower total volume, lower free-gel volume and higher concentration,
with similar (P>0.05) total number of spermatozoa, sperm kinetics and

REVISÃO DE LITERATURA
O GnRH é produzido em padrões pulsáteis pelo hipotálamo, que por
neurônios hipotalâmicos atinge a glândula hipófise e promove a produção dos
hormônios folículo estimulante (FSH) e luteinizante (LH). O FSH apresenta
receptores nas células de Sertoli, estimulando a gametogênese. Já o hormônio
LH atua nas células de Leydig, induzindo a esteroidogênese (andrógenos e
estrógenos). A produção do hormônio testosterona pelas células de Leydig é
fundamental para o desenvolvimento das características físicas e
comportamentais do macho devido as suas funções anabólicas e androgênicas
(AMANN, 1993; ROSER, 2001; ROSER, 2008).

efetuar a monta, obstruções em região de colículos seminais ou ducto deferente, azoospermia
e ejaculação retrógrada. O diagnóstico diferencial entre causas obstrutivas e azoospermia é
realizado pela dosagem da enzima fosfatase alcalina associada ultrassonografia transretal das
glândulas sexuais anexas. Já o diagnóstico de dor ou instabilidade durante a monta requer
uma avaliação clínica minuciosa geralmente associada a exames complementares. A
ejaculação retrógrada é diagnostica por sondagem vesical para recuperação da urina e

7.1.3. Ocitocina
1994, CARD et al., 1997, JOHNSTON and DeLUCA, 1998, DUTRA, 2000).
A indução da ejaculação utilizando detomidina também resultou em
ejaculados com menor volume total, maior concentração e maior número de
espermatozoides no ejaculado (ROWLEY et al., 1999, MRACKOVA et al.,
2013). Por outro lado, a utilização somente de xilazina resultou em parâmetros
seminais similares aos observados em coletas por meio de vagina artificial
(McDONNELL;LOVE, 1992).
A avaliação da morfologia espermática não resultou em alterações
significativas entre ejaculados provenientes de indução farmacológica e
ejaculados obtidos por coleta em vagina artificial, independente do protocolo
utilizado (CARD et al., 1997, DUTRA, 2000; NAOMI et al., 2012;
CORREA;BOZZO, 2016). Do mesmo modo, a avaliação das motilidades total e
progressiva não diferiram quando comparados os ejaculados obtidos por meio
de coleta em vagina artificial e por meio de protocolos de indução
farmacológica da ejaculação (MCDONNELL, 1991, MCDONNELL;ODIAN,
1994; MCDONNELL; TURNER, 1994, CARD et al., 1997, JOHNSTON and
DeLUCA, 1998, DUTRA, 2000; CORREA;BOZZO, 2016).

total number of spermatozoa, membrane integrity, sperm kinetics and
morphology compared to in copula ejaculates and xylazine protocols. The
semen characteristics suggests decreased accessory glands fluids in protocols
DO and IDO, probably resulting from the use of oxytocin, which increase the
epididymal tail contraction. Thus, oxytocin appears to play a role to induce
ejaculation when associated with detomidine, but not when associated with
xylazine.

Contrary, detomidine induced ejaculation with lower total volume, higher sperm
concentration and higher total number of sperm compared to in copula
ejaculates [4].
Chemical ejaculation presents a variable results regarding to time of
ejaculation depending on the drugs used. Most ejaculation occur as the animal
is becoming sedated, within 1-5 min after xylazine injection or as it is recovering
from sedation within 15-45 min [1-3,11-13]. Intramuscularly PGF2α treatment
resulted in ejaculations between 10-50 min [8] as well as treatments with
intramuscular detomidine resulted in ejaculation between 17-47 min.
The aims of the study were: 1) Compare the efficiency of different
protocols in inducing ex copula ejaculation; 2) To evaluate the success rates in
inducing ex copula ejaculation when oxytocin was added to the protocols;
3) Compare seminal parameters between in copula and in ex copula ejaculates.
This study was approved by the Animal Care and Use Committee, São
Paulo State University – UNESP Botucatu, SP, Brazil.
2.3 Trials
In the week before the start of the study, stallions were collected twice
with 2 days interval and then two semen samples were collected in a Botucatu®
model artificial vagina (Botupharma LTDA, Botucatu, SP, Brazil) from each
stallion using a phantom mount to obtain baseline measures. Induced
ejaculates were collected into a plastic bag held by a plastic ring positioned over
the prepuce and attached in the stallion’s flank by a tiny hope.
mL. This aliquot was used to examine sperm kinetic (IVOS version 12 Hamilton
Thorne Research, MA, USA) of each ejaculate and compared in copula
ejaculates and ex copula ejaculates. The CASA setup is described in Table 1.
The following parameters were evaluated: total motility (TM, %), progressive
motility (PM, %), angular progressive velocity (VAP, µm/s), progressive velocity
(VSL, µm/s), curvilinear velocity (VCL, µm/s) and percentage of rapid sperm
(RAP, %).

stallions in 70% of the trials. None of the stallions that responded to xylazine
treatments responded to detomidine treatments.
Interval from xylazine treatment to ejaculation ranged from 2 to 4 min in 3
stallions and to 15 to 20 min in 1 stallion. Interval from detomidine treatment to
ejaculation ranged from 2 to 7 min. No differences on the mean time of
ejaculation were observed when imipramine or oxytocin was associated to the
protocolos.

4. Discussion
sialorreha. Other studies administered imipramine orally crushed the tablets and
gave with feed [12], molasses [6] or sweet flavor [5] and did not report
sialorreha. Thus, similar side effects were described when imipramine
intravenous was used [13], evidencing that this effects it is probably due to an
cholinergic mechanism of action that justify this effect, since imipramine is a
tricyclic antidepressive with several unspecific mechanism of action.
In conclusion, the protocols tested did not induce ejaculation in stallions
above 4 years old. Also, this study observed that anxious and scared stallions
required higher doses to induce ejaculation. Thus, administration of the
protocols in the animal’s stall with a quiet environment was fundamental for the
success of the treatment. In the mean time, the use of different protocols with
detomidine and xylazine in the same stallion increased the success rates since
stallions that ejaculated with detomidine did not ejaculate with xylazine. Also,
oxytocin showed to increase the ejaculatory function associated to detomidine
with no adverse side effects.
