– I=0 These nuclei have no interaction with
the magnetic field and are therefore not visible
in a NMR. These nuclei have an even number of
protons and electrons, such as 12C, 16O and 32S.
– I = ½ These nuclei have a magnetic moment
and are visible in a NMR. Mainly 1H and 13C are
used.
– I>½ These nuclei are difficult to observe in
a NMR. Examples are 2H, 14N and 79Br.

Magnetic Shielding
• If all protons absorbed the same amount
of energy in a given magnetic field, not
much information could be obtained.
• Protons are surrounded by electrons that
shield them from the external field
(Shielded protons). Circulating electrons
create an induced magnetic field that
opposes the external magnetic field.
• Magnetic field strength must be increased
for a shielded proton to flip at the same
frequency.

Chuẩn bị mẫu
• Mẫu đo NMR thường là dung dịch
• Điều kiện cho dung môi:
– Hòa tan mẫu
– Không phản ứng với mẫu (No strong sample -
solvent interactions)
• Dung môi trong NMR thường không chứa
proton.
– Thường dùng CCl4 hay các dung môi trong đó
thay thế proton bằng deuterium (Deuterated
solvent).

• Độ dịch chuyển (chemical shift) thể hiện
mức độ chắn (shielding) trên proton
• Các yếu tố ảnh hưởng mức độ chắn
(shielding) trên proton:
– Electronegativity: Electrons are withdrawn by
neighbouring electronegative atoms
– Hybridisation: Protons attracted to sp2
carbon atom are less shielded
– Hydrogen bonding: Shielding is reduced by
hydrogen bonding

1. H-bonding increases with concentration
Common NMR solvents such as CDCl3 and CCl4
do not form H-bonds. When a sample
containing OH or NH groups is dissolved in
these solvents, the chemical shift of the OH
or NH protons changes with the concentration
of the sample.

2. H-bonding decreases with temperature
When a sample containing OH or NH groups is
heated, H-bonding is disrupted, and as a result
the chemical shift of the OH and NH protons
changes with temperature

3. H-bonding may occur between sample and
solvent
While CDCl3 and CCl4 do not form H-bonds to
OH or NH in a sample, water do. Such H-bonds
result in downfield shifts. In particular, the
position of water (a common impurity peak) is
strongly affected.
