NC = NSS 1. Prepare and label 2 tubes 3. Add 1 drop of Blood Sample on each portion P = POSITIVE 4. Mix using applicator stick N =NEGATIVE
5. Observe the result macroscopically 2. Add 2 drops of 5% RCS
6. Interpretation: 3. Add 2 drops of reagent:
w/in 2 mins P = D H2O
(+) Positive = Agglutination N = NSS (-) Negative = No Agglutination 4. Cover w/ parafilm and mix TUBE METHOD 5. Centrifuge for 15 secs at 3400 rpm 1. Prepare and label 2 tubes 6. Interpretation U = Undiluted D = Diluted No Hemolysis = intact cell button & clear supernatant 2. Add 2 drops of reagent Partial Hemolysis = Presence of cell button & pink supernatant U = Anti D Complete Hemolysis = absence of cell D = 1 drop anti D + 1 drop NSS button & red supernatant 3. Add 1 drop of 5% RCS
