1. Prepare a glass slide label as follows:

2. Add 1 drop of reagent in each portion:

 U = Anti D DEMONSTRATION OF HEMOLYSIS


 NC = NSS
1. Prepare and label 2 tubes
3. Add 1 drop of Blood Sample on each portion
 P = POSITIVE
4. Mix using applicator stick  N =NEGATIVE

5. Observe the result macroscopically 2. Add 2 drops of 5% RCS

6. Interpretation: 3. Add 2 drops of reagent:

 w/in 2 mins  P = D H2O


 (+) Positive = Agglutination  N = NSS
 (-) Negative = No Agglutination
4. Cover w/ parafilm and mix
TUBE METHOD
5. Centrifuge for 15 secs at 3400 rpm
1. Prepare and label 2 tubes
6. Interpretation
 U = Undiluted
 D = Diluted  No Hemolysis = intact cell button & clear
supernatant
2. Add 2 drops of reagent  Partial Hemolysis = Presence of cell
button & pink supernatant
 U = Anti D
 Complete Hemolysis = absence of cell
 D = 1 drop anti D + 1 drop NSS
button & red supernatant
3. Add 1 drop of 5% RCS
