langsam zugeben.
Reagentien:
33 % - NaOH Lösung:
500 g NaOH - Plätzchen werden in 1 Liter Deionat aufgelöst (starke Wärme-
entwicklung!)
2 % - Borsäure:
20 g Borsäure p.A. in 1l Deionat lösen.
0,1 N HCl:
Titrisol - Ampullen der Fa. Merck. Für 0,01 N HCl 0,1 N Lösung 1:10 verdünnen.
Standardlösung:
75 mg/l Glycin entsprechen 14 mg N org./l.
Nitrogen (NTOT according to Kjeledahl) (3 samples in duplicate)
Sample: dried (105 °C) and ground < 0.5mm (< 1mm if smaller is not possible)
Reagents:
H2SO4 (96 %)
Catalyst:
There are two catalysts which can be used for this method - Selenium reagent
mixture (toxic) or Kjeldahl tablets (Missouri catalyst - K 2SO4, Na2SO4, CuSO4).
33 % - NaOH solution:
500 g NaOH is disolved in 1l H2O deionised (attention: very high temperatures ocur!)
2 % - boron acid:
20 g boron acid p.A. are solved in 1l H 2O deionised.
0.1 N HCl:
Titrisol - from Merck 0.1 N HCl.
Standard solution:
75 mg/l Glycin which is 14 mg Norg./l.
Digestion:
1 g sample (dried, ground) accurate to 0.01 g resp. 20-50 ml liquid sample is put into a
Kjeldahl glas tube. A catalyst (1 tablet Missouri or 1.7 mg selenium reagent) and 15 ml
of the concentrated H2SO4 (95 %) are added. During the first hour the temperature is
raised up to 400 °C. At this temperature the glas tubes remain for 3 hours. Digestion is
completed, when receiving a white milky-cloudy colour. After cooling to room
temperature the distillation is done.
For liquid samples, the temperature is raised progressively; we need to pre-heat the
cold block to 55-60 °C before putting the flasks because of the exothermic reaction
(liquid sample + sulphuric acid). Within the first hour the temperature is raised to 160
°C, then to 230-240 °C and finally to 400 °C (total 4 hours). It is required to work in a
fume hood because poisonous and stifling gases are emitted. Digestion is completed
when receiving a clear liquid. After cooling to room temperature the distillation is done.
Distillation:
Then titration with HCl 0.1N (also other concentrations may be used, but then we have
observe this for calculating the results; when for example 0.01N HCl is used, a 10 times
higher amount will needed for titration. For calculation factor 10 has to be kept in mind).
The colour or boron acid is again getting red.
0.14007 * (amount of HCl) * 100
NKj [% DM] =
sample-weightdry* (100 – cont. of Resid. Water)
Always 2 replicates are measured. If the difference of one replicate to the average of
both replicates is more than 2.5 %, we have to repeat the analysis.
