PRINCIPIO: Cada vez que un antígeno ingresa en el cuerpo, el mecanismo protector de nuestro
cuerpo (Anticuerpo) comienza a protegernos y la inflamación es la respuesta de ese evento.
Podemos decir que esa es la parte del mecanismo de defensa del huésped. Hay varios factores
tisulares que desempeñan un papel activo e importante en el mecanismo de la inflamación.
Estos incluyen la liberación de histamina, bradiquinina y prostaglandinas. La inflamación
muestra el siguiente signo característico: I Tetón-Dolor-Enrojecimiento-Hinchazón.
Requisitos-
3. Equipo- Pletismómetro. Es un aparato simple que tiene dos tubos, de los cuales un tubo
tiene mercurio, y el otro tubo muestra el movimiento o la expansión de mercurio después de
colocar la longitud fija de la pata de rata. Cuando se coloca la pata, el nivel de mercurio
aumenta debido a la presión de desplazamiento aplicada por la pata de rata. Esto es mínimo
antes de la inflamación que aumenta después de la inflamación y luego se reduce nuevamente
cuando se administra el medicamento.
Procedimiento-
1. Primero pesa todas las ratas y numéralas adecuadamente en su cola. (en la base)
2. Haga una marca en la pata trasera de la rata experimental, desde 2 cm por encima de la
pata. Se hace solo para asegurar la correcta y misma inmersión de la pata cada vez.
3. Anote el volumen inicial de la pata de la pata izquierda y derecha de la rata solo con
observar el nivel de mercurio desplazado.
7. La pata derecha se mantendrá tal como está porque se la tratará como referencia para la
pata no inflamada para comparación.
8. Note el volumen de pata de ambas patas de control y de rata tratada con indometacina
como 15, 30, 60 y 120 min. después de la carragenina. 9. Calcule el% de diferencia tanto en la
pata de la rata de control como en el grupo tratado con indometacina según la reducción del
edema.
inflammatory Activity
PRINCIPLE- Whenever any antigen enters into body, our body's protective mechanism
(Antibody) starts protecting us, and the inflammation is response of that event. We can say
that is the part of host defense mechanism. There arc several tissue factor which plays active
and important role in mechanism of inflammation. These include- release of histamine,
bradykinin and prostaglandins. Inflammation shows following characteristic sign- I Teat-Pain-
Redness- Swelling.
Agent which give relief from inflammation are called anti-inflammatory drug. They work by
inhibiting the release of Histamine, and blocking the conversion of Prostaglandin into COX,
Specially COX-II.
The inflammatory reaction is rapidly produced in rat in the form of paw oedema, by utilizing
the stimulus or irritating agent like- carrageenan, fonualin solution, bradykinin, histamine,
mustered or egg white. When these agent given as injection in the paw of rat, sudden swelling
occurs, because of the release of histamine, 5HT, bradykinin and prostaglandins.
Requirements-
2. Drug- Carrageenan by preparing 1% w/v solution and by injecting its 0,1 ml to the plantar
region of rat. Indomethacin- 20 mg/kg by Sc route.
3. Equipment- Plethysmometer. It is a simple apparatus having two tube out of which one tube
is having mercury on it, an the other tube shows the movement or expansion of mercury after
placing the fixed length of rat paw. When paw is placed the level of mercury increased due to
displacement pressure applied by rat paw. This is minimum before inflammation which
increase after inflammation an then again reduce when drug is given.
Procedure-
1. Firstly weigh all the rat and number them accordingly on their tail. (at the base)
2. Make a mark on both the hind paw of the experimental rat, from 2 cm above the paw. Its
done just to ensure the proper and same dipping of paw each time.
3. Note down the initial paw volume of left and right paw of rat just by observing the level of
displaced mercury.
5. In one group we will inject simple saline and in another group we will inject our drug of
study that is indomethacin subcutaneously (SC)
6. After 30 min. inject 0.1 ml of carrageenan in the left paw of rat and it will treated as control
group.
7. The right paw will kept as it is because it will treated as reference for non inflamed paw for
comparison.
8. Note the paw volume of both legs of control and indomethacin treated rat as 15, 30, 60 and
120 min. after carrageenan. 9. Calculate the % difference in both paw of rat of control and
indomethacin treated group as per the reduction of oedema.
