THE INFLUENCE OF AGE AGAINTS INTERLEUKIN-1β PRODUCTION

INDUCED BY
Escherichia coli , Mycobacterium tuberculosis AND Streptococcus pneumoniae

Effectiveness of immunity dependent by coordination between innate and adaptive immunity,


which interacting use cytokine as mediator. Cytokines have a vital function in innate and adaptive
immunity. One kind of cytokine is Interleukin-1β (IL-1β), which secreted by macrophages,
monocyte, neutrophil, epithelial cells, NK cells, and T and B Cells. IL-1β regulate the mechanism
of cellular immunity to proctect from bacteri. Immune system, depending by many factors, one
is age. In general, human with older ages (≥65 years), will be through of decreasing immunity
component, like cytokine production. The aim of the study is to know the production level of IL-
1β in different ages. The sample of research is whole blood from human health (n = 946), whose
contact with tuberculosis patient and never has gotten medical of TB since 6 months ago. Each
is classified into 3 groups based on age (≤ 14 years, 15-64 years, ≥65 years). Whole Blood
Stimulation Assay (WBSA) uses culture method for this research. Whole blood from each group
was induced with E. coli, S. pneumoniae, and M.tuberculosis as stimulat IL-1β production. The
level of IL-1β production from supernatant of WBSA was measured by ELISA. Statistical analysis
conducted ANOVA, LSD with tukey. The result is shown increasing of IL-1β production
compared with control. The highest IL-1β production is stimulated by E.coli, second S.
pneumonia, the last M.tuberculosis. There is not different significance (p >0.05) of IL-1β
production in each age group. In this study there is evidence that in woman 40-60 years, there
is fluctuation of IL-1β production guessing of hormones, which explore with another study.
