Derivative Spectra

Spectral information is often masked by overlapping


absorption bands, sloping background absorption, or
the occurrence of a small peak or shoulder on the side
of a broad absorption band. Derivative spectroscopy
may be used to enhance fine detail, reduce the effects
of a sloping background and accurately locate a masked
band for measurement.

Derivatives may be displayed alone or superimposed


on the fundamental, with or without an offset.

Band Quantification

For accurate quantitative work it is often required to


determine the area or height of an absorption band
corrected for background absorption.

A very easy to use programme is provided for this


purpose. The analytical result is displayed on the
screen, with construction lines, enabling the quality of
the chosen parameters to be assessed.

Band quantification may be stored as a method

Multi-Cell Kinetics
The Super Aurius Series offers multi-cell Kinetics as
well as single cell Kinetics as available on the Aurius
Series. Plots are displayed on the screen and printed.

Regression analysis is applied to the user selected

FAST DATA TRANSFER TO PC

DataStream
Cecil DataStream provides rapid transfer of
raw data, or data processed and formatted
by the spectrophotometer, to a PC for
instant display. Scans, measurements,
programmed measurements and
sophisticated analytical procedures may all
be transferred with great ease.
Transferred data may be saved on disk or
incorporated into a spread sheet or maths
pack. Incorporation into Microsoft Excel is
shown here for display, manipulation,
graphical presentation, archiving or report
generation. Full PC control of the
instrument is provided.
Simplicity of operation is a key feature as
any familiar spreadsheet or maths package
- Excel, Lotus 123, Grafit etc - may be used,
avoiding a learning cycle and saving
valuable time. Reports are printed using
any printer supported by Windows.

Advanced Kinetics
For more advanced reaction kinetics data
manipulation, raw kinetics data generated
by the spectrophotometer may be captured
and transferred by DataStream into Grafit
Software.
Using a suitable template the Lineweaver
Burk transformation, shown here, was
automatically plotted and the values of Km
and Vmax extracted. Other transformations
such as Eadie-Hofstee etc. may be used.
Linear, non linear, or polynomial curve
fitting are easily performed together with
linear regression analysis.

Absorbance Validation
The Pharmacopoeia (EP/BP) test for absorbance
accuracy is one of the tests provided. The test
measures the values of peaks and valleys in the
absorbance spectrum of a 60mg/L solution of
Potassium Dichromate in a 10mm pathlength cuvette.
The results of such a test run are shown here.

Validation Using Filters

Absorbance and Wavelength validation are most


conveniently carried out using filter sets calibrated by
Cecil Instruments on its reference spectrophotometer
against standards certified by the NPL. Full validation
programmes for these tests are provided.

Straylight

Straylight may be measured using the Pharmacopoeia


(EP/BP) test shown here. This test uses a 1.2% W/V
solution of Potassium Chloride in water, measured
against water in the reference beam. An Absorbance in
excess of 2A must be measured at a wavelength of
200nm.

The ASTM tests at 220nm and 340nm are more widely


used and are provided.

Derivative Spectra

Spectral information is often masked by overlapping


absorption bands, sloping background absorption, or
the occurrence of a small peak or shoulder on the side
of a broad absorption band. Derivative spectroscopy
may be used to enhance fine detail, reduce the effects
of a sloping background and accurately locate a masked
band for measurement.

Derivatives may be displayed alone or superimposed


on the fundamental, with or without an offset.

Band Quantification

For accurate quantitative work it is often required to


determine the area or height of an absorption band
corrected for background absorption.

A very easy to use programme is provided for this


purpose. The analytical result is displayed on the
screen, with construction lines, enabling the quality of
the chosen parameters to be assessed.

Band quantification may be stored as a method

Multi-Cell Kinetics
The Super Aurius Series offers multi-cell Kinetics as
well as single cell Kinetics as available on the Aurius
Series. Plots are displayed on the screen and printed.

Regression analysis is applied to the user selected

DataStream
Cecil DataStream provides rapid transfer of
raw data, or data processed and formatted
by the spectrophotometer, to a PC for
instant display. Scans, measurements,
programmed measurements and
sophisticated analytical procedures may all
be transferred with great ease.
Transferred data may be saved on disk or
incorporated into a spread sheet or maths
pack. Incorporation into Microsoft Excel is
shown here for display, manipulation,
graphical presentation, archiving or report
generation. Full PC control of the
instrument is provided.
Simplicity of operation is a key feature as
any familiar spreadsheet or maths package
- Excel, Lotus 123, Grafit etc - may be used,
avoiding a learning cycle and saving
valuable time. Reports are printed using
any printer supported by Windows.

Advanced Kinetics
For more advanced reaction kinetics data
manipulation, raw kinetics data generated
by the spectrophotometer may be captured
and transferred by DataStream into Grafit
Software.
Using a suitable template the Lineweaver
Burk transformation, shown here, was
automatically plotted and the values of Km
and Vmax extracted. Other transformations
such as Eadie-Hofstee etc. may be used.
Linear, non linear, or polynomial curve
fitting are easily performed together with
linear regression analysis.

Absorbance Validation
The Pharmacopoeia (EP/BP) test for absorbance
accuracy is one of the tests provided. The test
measures the values of peaks and valleys in the
absorbance spectrum of a 60mg/L solution of
Potassium Dichromate in a 10mm pathlength cuvette.
The results of such a test run are shown here.

Validation Using Filters

Absorbance and Wavelength validation are most


conveniently carried out using filter sets calibrated by
Cecil Instruments on its reference spectrophotometer
against standards certified by the NPL. Full validation
programmes for these tests are provided.

Straylight

Straylight may be measured using the Pharmacopoeia


(EP/BP) test shown here. This test uses a 1.2% W/V
solution of Potassium Chloride in water, measured
against water in the reference beam. An Absorbance in
excess of 2A must be measured at a wavelength of
200nm.

The ASTM tests at 220nm and 340nm are more widely


used and are provided.

PROGRAMMED FOOD AND
DietQuest ™ for Food and Beverage Analysis
Cecil Instruments offer three DietQuest™ spectrophotometers
fully pre-programmed for both food and beverage analysis.
Methods include single and multi-wavelength measurements
and spectral scanning. The DietQuest™ UV and the VIS
versions are as shown on the right.
All the pre-programmed methods are defined by national and
international bodies and trade associations including the
following:

AACC, AOAC, ALVA, APA, ASBC, BS-EN, CAA, EBC, DIN, FDA,
FIL-IDF, GUdRI, ICC, IFU, IoB, IOCCC, ISO, MAFF, MEBAFA,
MEBAK, NEN, NF, OIV, OJEC.

Simple Rapid Operation


Even the most complex assay is carried out simply by

Reaction Kinetics
Enzyme reaction methods using kits supplied by Boehringer
- Mannheim, Roche, Diogen, Megazyme etc. have been pre-
programmed for a single cell or for 4 or 6 cells in an
automatic changer.
Reaction curves are displayed on screen and regression
analysis applied to the user selected part of the curve. A
number of the assays are simple end point measurements.
Sample temperature control, where required, is available
using either water circulation or Peltier Thermoelectric cell
holders and controller.

DataStream - Data to a PC
Both raw, and processed and formatted data may be rapidly
exported to a PC for incorporation into a spreadsheet or
mathematical package of the users choice, e.g. Excel, Grafit
etc. for manipulation and report generation.

Samples of 300µl or less, and multiple samples, may be
measured with very low cross contamination.

Samples are measured in the same cell enhancing


accuracy and speed of measurement. The pump, in the
sample compartment, is controlled by the instrument's
microprocessor.

Ultra-Micro Sampling
Valuable samples, such as biological samples prepared
with costly reagents, may be measured using a special
ultra-microcell and holder. With the system, samples of
50µl or even less may be accurately measured.

Tablet Dissolution
The CE 7460 and CE 7560 versions of Aquarius are fitted
with a special cell compartment and pipe port for tablet
dissolution assays.

The instrument may be used with either a six or eight

for mathematical interpolation and calculation. In this particular example, the basic data plot can yield the limiting
rate (Vmax), the Michaelis constant (Km) and the initial linear portion (Vmax/Km).

This has been transformed into the Eadie-Hofstee format and plotted to give better precision in the calculation of
these important factors and constants.

Other transformations are available such as Lineweaver-Burk and the Hill plot for allosteric enzyme systems.

Lambda Max Scans
The same trial mixture as before, which separated into
three peaks under isocratic conditions, separated into four
peaks under binary gradient conditions using an Adept
System 4S.

Each peak was scanned at its maximum - Lambda max scan


– and the spectra displayed whilst the chromatogram was
simultaneously displayed and quantified by integration to
provide baseline corrected peak areas and peak heights.

Peaks 1, 2 and 4 are quantitatively similar and all have peak


maxima close to 222 and 273nm.

Peak 3 scan is clearly very different having maxima near


234, 268, 316, and 331nm.

Optimum monitoring wavelengths for each peak may be


chosen from these scans.

The photo shows an Adept Isocratic System 2S. The


WaveQuest detector, fitted with a column and injector valve
panel, is mounted on the CE4100 pump, with the
PowerStream interface unit at the bottom of the stack.

An optional column oven is fitted on the right. The


system offers Diode Array performance and the very best
dual and variable wavelength monitoring, with
exceptional compactness, as does the Adept Gradient
System 4.

Normalised Overlays Confirm Purity
Automatic peak purity checks were made by the same
upside and downside, “on-the-fly”, scans for all four
peaks.

The results are shown for peaks 3 and 4 only, overlayed


before and after normalisation of the spectral scans. Peak
4 is shown in the upper two plots and Peak 3 in the lower
two.

From the overlays, particularly when normalised, it is


seen that a near perfect match is obtained indicating that
these peaks were homogenous and hence pure, as also
were Peaks 1 and 2.

Eight Scans of Peaks


Several means of triggering the wavelength scan of a
peak are user selectable.

In the example shown here, WaveQuest was


programmed to make repetitive scans of pyrene as it
eluted.

The use of the cursor is demonstrated by using it to


measure the absorbance maximum at 326.7nm. The drop
line shows that all the scans have the same wavelength
for this absorption maxim.

Spectral Overlay Confirms Identity
A different mixture of the same components was used to
produce the chromatogram shown here with its 4 peaks.

Spectral library searches were made in order to identify


the components, in conjunction with retention time data.

Peaks 1, 2 and 4 can be identified as alkyl phthalates, most


probably dimethyl, dibutyl and di(ethylhexyl) phthalates.

Analysis of Sugar Free Soft Drinks
The new ultra fast scanning detector from Cecil
Instruments, the WaveQuest, was used as part of an
Adept System 4S to separate, identify and quantify the
components of soft drinks.

Using the CE4300, high quality UV-Visible spectra were


obtained “on-the-fly” without the use of an expensive

were injected and eluted under the same conditions as
the sample mixture.

positive identification of all the components.

DATA PROCESSING + CONTROL
Pump-Proven Performance

The Cecil CE4100 dual piston solvent delivery pump is


well established as very reliable, convenient to use,
with a very high performance specification.
The microprocessor controlled pump uses a precise
stepping motor drive for the dual overlapping pistons
which deliver solvent with very little pulsing.
Electronic feedback is used to control the pump speed
during its cycle to reduce pulsing to a negligible level.
No further mechanical pulse damping is required.
Delivery of mobile phase in the range 0.2 – 2.5mL per
minute, the usually used range, is well within the

Convenient Column Heater/Chiller (Oven)


It is important that the separation columns should operate at a
constant temperature, matching the temperature of the measuring cell.

The column oven module is a very convenient front


opening design accommodating two analytical columns and pre-
columns and also, when needed, a mobile phase background
conductivity suppressor cartridge and its over-pressure protection
valve.

The oven operates over the range from 5 ºC to 70 ºC


(maximum 12 ºC below ambient).

The default temperature is set at 35ºC.

The actual and set temperatures are displayed by a bright LCD


display and may be controlled by the adjacent keypad, or set by the
PowerStream software. Temperature stability is better than ±0.01ºC.

The flow tubes from the columns and suppressor cartridge enter and
leave the oven enclosure through convenient self-sealing ports. The
output port to the cell thermostatted enclosure lines up with the input
connector of the enclosure, when the modular system is arranged as
shown in the photographs of the system.

The very close coupling to the detector cell leaves only about 2
centimetres of tube between the oven and the cell. With the cell and
oven operated at the same temperature, say 35ºC, drift or noise due
to ambient fluctuations is eliminated.

A column switching valve may be fitted to the oven offering


convenient operation.

WITH HIGH SENSITIVITY
Phosphoric Acid in Soft Drinks

The established method for


determination of Phosphoric Acid in
cola beverages uses the
“molybdenum blue” analysis. This
spectrophotometric method is not
however rapid.

An Ion Chromatography method was


developed, in our applications
laboratory, which is both rapid and
accurate. The Cecil CE4715
suppressor was used to reduce
background conductivity. This
improved sensitivity with reduced
noise and drift. The reduction was
from about 500µS/cm to 20µS/cm.

The flow rate of the eluent: 2 mM


Na 2CO3 / NaHCO 3 buffer, was
1.3mL/min with a 7 micron anion
column.

The determination of Phosphoric acid in a cola drink, as shown here, The column oven and cell were both

CE 7260 Spectrophotometer
This symmetrical double beam spectrophotometer
from the Aquarius series, with an 8 cell changer, has a
1.8nm optical bandwidth and is ideal for dissolution
measurements. Its stability of, ±1 x 10-4A/hour,
approximately ten times better than single beam
instruments, makes it ideal for extended
measurements such as sustained release monitoring.
Full cell and wavelength programming together with
wavelength scanning make the instrument ideal for
dissolution applications and may, when required, be
used as a high quality scanning double beam
spectrophotometer for other applications.

Operation is simple following menus and prompts


displayed on the LCD screen, using the keypad to
interact with the comprehensive embedded software.
Validation software is available. Used in System 1.

CE 9260 Spectrophotometer
This is a top performance double beam spectro-
photometer from the Super Aquarius Series. The
optical system is fully symmetrical and uses a high
performance end window photomultiplier as detector.
This ensures the very best performance with minimum
drift and the best possible noise performance,
particularly for low energy conditions.

Both method and spectral scan storage are provided


together with powerful software for all applications.

For top performance this is the instrument of choice.


Validation software is available. Used in System 3.

CE 3021D Spectrophotometer
This is a high quality single beam spectrophotometer
from the Super Aurius series, with an 8 cell changer
and 1.8nm optical bandwidth. Its stability of 1 x 10-3
A/hour makes it suitable for dissolution monitoring,
and may be used as a general purpose or scanning
spectrophotometer if required.

Full wavelength and cell programming, as well as


wavelength scanning, are possible. Operation is via
embedded software using an LCD screen to display
menus, prompts etc. with entries made through its
comprehensive keypad. Enhanced accuracy is
provided, as with the CE 7260, by the very low, 0.01%,
straylight. Software for validation is available. Used in
System 2.

CE 3791 Peristaltic Pump


The CE 3791 is a multi-cartridge design with good flow
accuracy. Each of the eight channels may be individually
released, or adjusted for roller operating pressure.
