PROCESS TO MAKE MOTHER CULTURE
Materials:

 Potato (250g)  Alcohol lamp


 Distilled water (1 lit.)  Inoculation needle
 White or brown Sugar (3 tbsp.)  Blade
 White Gulaman bars (3 bars)  Cotton
 Good & fresh mushroom for culture  Aluminum foil
 Cracked Corn  Flat bottle
 4” x 11” x .03 PP bag  Alcohol

PROCEDURE FOR CULTURE MEDIA

 Boil the 250gpotato in the 1 lit. Distilled water and remove the potato after boiling.
 Add 3 tbsp. sugar and the 3 barsgulaman bars while boiling mix well.
 After boiling about 5 min. transfer the cooked PDA to the flat bottle with about 40ml.
 Sealed the bottle with cotton plug & aluminum foil and sterilize it in pressure cooker with about 20
min. starting from its boiling pressure.
 Let the flat bottle place sided and cool off.
 After coolingdown,wipe the mushroom with alcohol and slice a small piece tissue in the center from
the young mushroom
 Put the tissue inside the bottle with PDA at the center
 Sealed the PDA in the bottle with clean cotton.
 After 2weeks mother culture is ready for transferring to grain spawn
 Boil the cracked corn for about at least 5 min., mix well while boiling.
 Drain and dry the cracked corn for about 60% moist.
 Packed the cracked corn with about 250g in the PP bag.
 Sterilize it in pressure cooker with about 20 min. starting from its boiling pressure.
 Transfer the culture media to the grain spawn with just a small amount by the process of heating the
bottle mouth before and after transferring.
 After 2-3 weeksGrain spawn is already ramified and ready for inoculation

Note:
 Must use clean chamber for processing
 Must be sanitized hands, no accessories and clean nails.
 Must be medium fire only for sterilizing.
 Don’t open the pressure cooker after sterilizing the bottle it might blow the bottle, cool down
first.
 Make sure the cotton plug must be extended from the mouth of the bottle for the holder.
 Remove water from the PDA after sterilizing and before transferring tissue.
 Don’t hold the cotton plug of mother culture when transferring the culture media to grain
spawn.
 Make sure when transferring culture media it’s not too much away from the fire
 1 Mother Culture bottle can produce 30 packed 250gGrain spawn
 1 lit. of Distilled water can produce 18 bottle of Mother Culture

Incubation at pamumungasa Fruiting Bags

Ingredients:

Oyster mushroom (you can have as much as you want)


1 cup cornstarch
2 tbsp salt
2 tbsp powdered garlic
Dash of ground pepper
