How to perform Interference testing
Bilirubin
1. Conjugated bilirubin
Test Material : Dissolve 20mg unconjugated bilirubin in 2mL 0.1N NaOH. (1000 mg/dL)
Test Pool A : Add 0.2mL of test material to 10mL pooled plasma with stirring. (20 mg/ dL)
Test Pool B : Add 0.2mL of 0.1N NaOH to 10mL pooled plasma with stirring.
Test Pool A and B are mixed as following,

Test Pool A : Add 0.2mL of test material to 10mL pooled plasma with stirring. (20 mg/ dL)
Test Pool B : Add 0.2mL of distilled water to 10mL pooled plasma.
Test Pool A and B are mixed as following,

Hemogulobin

Test Material : Hemolysate made by Osmotic shock procedure or Mechanical lysis procedure.
Test Pool A : Add measured volume of hemolysate to 10 mL pooled plasma to make 500
mg/dL hemogulobin.
Test Pool B : Add equivalent volume of saline to 10 mL pooled plasma.
Test Pool A and B are mixed as following,

How to perform Interference testing
Lipemia
Test Material : 20% fat emulsion (Intralipid, Cutter Laboratories)
Test Pool A : Add 0.5 mL of 20 % Intralipid to a 10 mL volumetric flask. Make up to
volume with pooled plasma.
Test Pool B : Add 0.5mL of distilled water to 10mL pooled plasma.

5. Lipemia
Test Material : 20% Intralipid, Cutter Laboratories
Test Pool A : Add 0.5 mL of 20 % Intralipid to a 10 mL volumetric flask. Make up to
volume with pooled plasma.
Test Pool B : Identical, except use 0.5 mL saline or Owren`s veronal buffer in place of
the lipemic stock.
