Operating procedure for INCUBATORS

 This is the standard operating procedure for all types and makes
of incubator used in the school of li8fe sciences.
 Also listed are all hazards and risks associated with the use of
incubators those persons at risk and precautions to be taken.
 Required controls for use of incubators.
 All containers placed in incubators should be labelled with the
name, date, and contents.
 Containers should be removed after the appropriate time of
incubation to prevent overcrowding or contamination of the
incubator.
 Any spillage should be reported immediately to the laboratory
technician and should be cleaned up in a manner appropriate to
the materials spilled.
 Where temperatures in excess of 370ºC are in use ,appropriate
gloves must be worn.
 Any faults associated with an incubator should be reported
immediately to the laboratory technician.
 If an incubator is faulty it should be switched off until repaired.
 Unauthorized personnel should not attempt to dismantle any
part of an incubator for any purpose.
 Instructions should be subjected to servicing based on the
manufacturers recommendations.
 Any cuts sustained should be dealt with by first aid personnel
and reported.
 Heat, electricity, chemicals, microorganisms, damged containers
are the hazards associated with incubators.
 Microbial contamination, clothing, burns to skin, electric shock
are the risks associated with incubators
Operating of LAMINAR FLOW
1. Turn OFF UV Light.

2. Carefully remove the front covers. (Store safely)

3. Turn on the fan (motor) for sterile air flow to commence.

4. Turn on the “light” for visibility (Not UV Light)

5. Swab the work surface with 70 % Ethanol

6. REMEMBER :

 Never leave a flame unattended (Turn off gas when you are

away from the cabinet)

 DO NOT have flammable liquids near the flame

 Flammable liquids must be in a glass container – (Not

Plastic)

 Gas Supply must when TURNED OFF when changing

cylinders and removing the regulator.

7. Perform tasks as required within the Laminar Flow using sterile

technique.

8. Upon completion remove any items from within the cabinet

9. Turn OFF UV Light.


10. Carefully remove the front covers. (Store safely)

Operating of SCANNING ELECTRON
MICROSCOPE
PROCEDURE
OPERATION :

 High Vacuum light is on and ACC voltage ready lamp is on and


not flickering, press ACC voltage ON/OFF switch. Press the up
arrow to increase.
 CRT screen will start in a raster mode. Press 􀀀 to switch to TV
mode.
 If instrument was left in working order. Pressing the auto then
ABC buttons a few times should bring the image into view.
 Switching to manual and adjusting the brightness and contrast
seems to work better than auto.
 As above, auto focus coarse and fine and auto Stigmator followed
by fine manual adjustment should bring a fairly clear image into
view.
 At this point one can increase the magnification adjusting the
above at each step.
 At some point the accelerating voltage may have to be increased,
the aperture adjusted, and C. Lens adjusted to obtain a clear image
that does not wander on the CRT too much.
 The operator should be trained in these adjustments and refer to the
manual for further details. Reducing the condenser lens current
excessively with the post HV = on (as in normal operation) can
cause too much secondary electron emission which may damage
the scintillator.
Operating of REFRIGERATOR
. RESPONSIBILITY
Operation and Calibration – Operator Compliance –
MICROBIOLOGY

4.PROCEDURE
OPERATION :
1. All refrigeration/freezer units should be regularly inspected and kept
clean.
2. Food and beverages for human consumption will only be stored in
refrigerator/freezers located in designated employee break rooms.
3. All reagents, biological samples, animal diets, carcasses, or any
substances requiring refrigeration will be placed in an appropriate
container to prevent the possibility of spills, leakage, odors, or spoilage
prior to storage in a refrigerator/freezer unit.
4. Bottles, tubes, and containers with a cap or top must be stored in an
upright position
and in such a way to insure their stability.
5. All reagents, biological samples, animal diets, carcasses, or any
substances stored
in a refrigerator will be appropriately labeled (with the exception of
carcasses/biohazardous waste being held for disposal). At a minimum
labels should include the contents, PI, IACUC protocol number, and
date. Additionally, containers of substances that have been removed
from their original manufacturer/distributor packaging must be labeled
so as to identify their contents and any health or safety precautions that
should be considered.
6. Canned foods and special animal diets once opened should be
appropriately covered and dated to ensure against contamination and
spoilage during storage.Perishable food items having been opened more
than 10 days will be discarded.
7. Reagents, animal diets, samples, containers or carcasses not properly
labeled will be considered the property of Comparative Medicine and
may be utilized or disposedof at the discretion of the Facility Manager.
8. Explosive, flammable, or corrosive substances are not to be stored in
generalpurposerefrigerator units and should be stored in laboratory
refrigerators specifically designed for that purpose.
EQUIPMENT MONITORING
1. The Surgical Core Manager and the Facility Manager are responsible
for the
monitoring of refrigerators and freezers located within animal facilities
and used for
the storage of reagents, pharmaceuticals, biological samples, animal
diets, or other
substances that require specific environmental temperature control and
could affect
the generation, measurement, or assessment of research data.
2. High and low temperature range will be recorded at least weekly on
the
Temperature Monitoring Log
located on the unit being monitored. This log should
contain the unit or ID number, the facility, and location of the unit, the
highest and
lowest temperature recorded, date, and initial of person doing the
monitoring.
3. At least monthly the Surgical Core Manager or Facility Manager shall
inspect, date,
and initial the log to insure the unit and its contents are being properly
maintained.
Monthly, these logs will be submitted to the Facility Manager for
retention in accordance

Operating of MELTING POINT APPARATUS

PROCEDURE
OPERATION :

4.1 Keep the ‘HEATER’ & ‘STIRRER’ Knobs to ‘MIN’.


4.2 Switch on the Main power supply. Switch on the ‘MAINS’ of
the instrument.
4.3 The display will show some random figure, then press the
‘RESET’ push button to display the actual temperature of the
furnace.
4.4 Keep the melting point tube in silicone oil containing glass
container. . Check through the lense type mirror about the capillary
is correctly seated in its cavity.
4.5 Turn the STIRRER knob slowly towards MAX. Press the
START push button.
4.6 Turn the heater knob to MAX and put HEATER HIGH/LOW
switch to HIGH. ( This switch is provided on right hand side of the
instrument ). The raise of temperature can be controlled by using
this switch and the HEATER knob.
4.7 When the temperature reaches within about 30 o C, of the
expected melting point, reduce the rate of heating and adjust the
rate of rise of temperature to about 1 o C, per minute.
4.8 Wait till the melting point is reached which is indicated by
sample is melted in the tube. The digital read-out will display the
exact temperature at which the sample is melted. Record this
temperature. Turn off the ’HEATER’ Knob.
4.9 After reducing the temperature, switched off the instrument.
