Assignment: Conduct the following activity to compare the structure of cheek cells and epidermal
cell of a red onion. You are provided with these materials: electronic microscope,
slide, specimen cover slip, blue methylene solution, toothpick, tissue paper, red
onion, razor blade, forceps and water.

Follow these instructions:

A. To observe the structure of cheek cells


1. Rinse your mouth. Using the blunt end of a clean toothpick, carefully scrape some cells
from the inside of your cheek.
2. Spread the scrapings onto a clean slide.
3. Add a drop of blue methylene solution to the smear.
4. Place a cover slip at an angle to the water on the slide. Slowly slide the cover slip to cover
the specimen without trapping any air bubble.
5. Clean and dry up any water on slide and around the cover slip with tissue paper.
6. Examine the cheek cells under the microscope. Draw and label the structure of a cheek cell.

B. To observe the structure of epidermal cells of a red onion


1. Cut a red onion into half with a razor blade. Cut into the middle part of the red onion till
the outer leaf.
2. Place a clean slide onto it. Remove the other leaves except the epidermal layer.
3. Place a drop of water onto the specimen.
4. Place a cover slip at an angle to the water on the slide. Slowly slide the cover slip to cover
the specimen without trapping any air bubble.
5. Clean and dry up any water on the slide and around the cover slip with tissue paper.
6. Examine the red onion cells under the microscope. Draw and label the structure of red
onion cell.
