1. Introduccin.
Dentro de este campo la presente memoria est dirigida al estudio de las reacciones
de acoplamiento cruzado catalizadas por metales de transicin (paladio y hierro),
as como el estudio de las reacciones de adicin de nuclefilos de oxgeno a
enlaces triples catalizadas por oro. A modo de introduccin se ha considerado
interesante describir brevemente las lneas fundamentales de la evolucin histrica
de estas reacciones.
Actualmente, estas reacciones pueden llevarse a cabo con una amplia variedad de
reactivos organometlicos y electrfilos, los cuales pueden agruparse en funcin
del metal del compuesto organometlico y del catalizador empleado. As pues, se

En contraste con el desarrollo actual, hasta comienzos de 1960, los nicos metales
para los que se conocan reacciones de formacin de enlaces carbono-carbono eran
el Li y el Mg.2 La elevada reactividad que caracteriza a estos compuestos
organometlicos, limitaba su utilidad sinttica a reacciones de acoplamiento
cruzado con slo determinados haluros de alquilo no impedidos (como pueden ser
los haluros de alquilo primarios, los de alilo y los de bencilo entre otros). Por otro
lado, los organometlicos de litio y magnesio se caracterizan por su
incompatibilidad con muchos grupos funcionales y por las reacciones competitivas
de intercambio halgeno-metal dando lugar a productos de homoacoplamiento.

Sin embargo, los sustratos ms frecuentes son los alquinos, Thomas24 describi la
sntesis de cetonas mediante la reaccin de hidratacin de un alquino terminal
catalizada por cido tetracloroaurico. En 1985 Hutchings estudi la reaccin de
hidroclorinacin del etino catalizada por oro soportado25, siendo ste un mtodo
alternativo al empleo de sales de mercurio. Estos estudios propiciaron el desarrollo
de la qumica del oro. Entre los primeros nuclefilos empleados en las reacciones
de adicin a alquinos se encuentran las aminas26 y los alcoholes27, en estas
reacciones los catalizadores de oro demostraron tener una eficacia superior al
paladio, si bien, no ha sido hasta 1998 y 2003 cuando se han llevado a cabo las
primeras reacciones de hidroalcoxilacin28 e hidroaminacin29 intermolecular,
respectivamente. Asimismo, en los ltimos aos se han empleado alquenos30,
cidos carboxlicos31, cetonas32, iminas33 e incluso sulfxidos34 como nuclefilos
tanto en reacciones de adicin intramoleculares como intermoleculares.
Por otro lado, el primer ejemplo de formacin de enlace C-C fue descrito en 1993
por Gasparrini.35 Este autor describi la sntesis de oxazoles mediante reaccin
entre dos alquinos y xido de nitrilo. Ya en el 2000 Hashmi describi las

arilacin de vinil sulfxidos por reaccin de un -halo vinil sulfxido con un cido
bornico como uno de los objetivos del primer captulo.
Las reacciones de adicin de nuclefilos a enlaces mltiples catalizadas por oro son
tambin un mtodo til para la formacin de enlaces C-C y C-heterotomo, debido
a su simplicidad experimental. Dentro de este campo, son muy frecuentes las
reacciones de adicin intramolecular a alquinos de alquenos y carbonilos, por el
contrario las reacciones intermoleculares son todava un campo en desarrollo. Por
ello, se consider interesante estudiar las reacciones de adicin intermolecular de
sulfxidos a alquinos como objetivo del segundo captulo.
Por otro lado, se plante como objetivo del tercer captulo una combinacin de las
reacciones de acoplamiento cruzado catalizadas por paladio y de las reacciones
de adicin de nuclefilos a un enlace mltiple en molculas con un grupo ter de
oxima. Sealar que esta ruta sinttica presenta una serie de ventajas; en primer
lugar, el ter de oxima proparglico obtenido a travs de una reaccin acoplamiento
cruzado de Sonogashira da entrada a la obtencin de aminas homoproparglicas. En

segundo lugar, las cetonas obtenidas en las reacciones de hidratacin de los teres
de oxima proparglicos (alquino interno) presentan una regioqumica bien definida,
a diferencia de las mezclas de regioismeros obtenidas habitualmente.

Aunque la reaccin tiene lugar con variedad de cidos bornicos, los mejores
resultados se obtienen cuando stos poseen sustituyentes dadores de electrones en
el anillo aromtico. De hecho, sustituyentes con tomos de oxgeno o flor en
posicin orto, es decir, con una elevada capacidad de coordinacin en la esfera del
metal ralentizan la etapa de eliminacin reductora. Por otro lado, la selectividad
observada en la reaccin de carbonilacin frente al acoplamiento cruzado directo
no poda explicarse admitiendo que la reaccin transcurra a travs de un ciclo
cataltico convencional. Por el contrario si era coherente si se admite que la
insercin del monxido de carbono tiene lugar tras la etapa de transmetalacin.

Por otro lado, Farina ha aplicado las reacciones de acoplamiento cruzado de Stille
entre un alquenil estannano y un alquenil triflato a la sntesis de inhibidores de -
lactamasa (esquema 1.5).11 Estas estructuras, de acuerdo con el concepto
introducido por Firestone y Walsh12, deban de generar un sulfxido allico en el

Aunque los rendimientos fueron elevados, el ZnCl2 era necesario para acelerar las
reacciones. En lo que se refiere a la estereoqumica de la reaccin, nicamente en
el caso de que R sea un grupo p-tolil se obtuvo un nico diasteroismero, con el
resto de los sustratos se produjo la racemizacin del producto de reaccin en todos
los casos.
En 1995 Paley13 utiliz los 2-halo vinil sulfxidos para la sntesis de eninos, a
travs de reacciones de acoplamiento cruzado de Sonogashira. Siendo sta una
metodologa bastante general para alquinos funcionalizados (esquema 1.6). La
presencia de una base, como la DBU, era necesaria para suprimir la reaccin
secundaria de adicin-eliminacin al doble enlace.

Los primeros ensayos realizados fueron con el ismero trans del sulfxido. Esta
reaccin demostr tener un carcter general, debido a los buenos rendimientos
obtenidos (mezclas racmicas Z/E: 80/20) con distintos vinil estannanos y vinil
sulfxidos utilizados (esquema 1.8).

Sin embargo, esta metodologa no era til con el ismero cis del sulfxido, debido
a una isomerizacin del doble enlace. Esta isomerizacin era resultado de la
adicin conjugada de PPh3 proveniente del sistema catalizador al vinil sulfxido,
seguida de una rotacin de este intermedio y salida de PPh3. Posiblemente, el
paladio coordinaba al oxgeno del sulfxido facilitando el ataque nucleoflico. Para
evitar este inconveniente la reaccin deba llevarse a cabo en ausencia de ligando
(esquema 1.9).

Por otra parte y dada la toxicidad de los reactivos de estao, otros autores han
utilizado otros reactivos organometlicos como cidos bornicos, derivados de cinc
y alquinos terminales para la sntesis de dienil sulfxidos mediante reacciones de
acoplamiento cruzado catalizadas por paladio.

En sus estudios Ma demostr que los 2-alquenil sulfxidos tambin podan sufrir
reacciones de acoplamiento cruzado de tipo Negishi o Sonogashira para la
obtencin de eninos.15 En el primer caso, se haca reaccionar el sulfxido con
PhZnBr en presencia de DMF (ecuacin 1, esquema 1.11). Mientras que en el
segundo caso, las reacciones tenan lugar con alquinos terminales catalizadas por
Pd(PPh3)2Cl2 y CuI en presencia de Et3N y DMSO como co-disolvente (ecuacin
2, esquema 1.11).

Aunque las reacciones con sales de sulfinatos no son una metodologa muy
extendida s que se han aplicado puntualmente para la sntesis de sustratos como
las bencil sulfonas y vinil sulfonas.

enlace C-O tena lugar a travs de la formacin de un intermedio de paladio
benclico (I, esquema 1.20). Un equilibrio entre los intermedios I y II permitira
explicar la diferente grado de reactividad de los sustratos con impedimento

-En el estudio de la reaccin se concluy que la conversin en el producto de
acoplamiento cruzado estaba altamente influenciada por la estructura del
catalizador de hierro(III). As pues, complejos de hierro (III) con ligandos del
tipo -dicetonas eran los ms efectivos, encontrndose entre los ms habituales

el Fe(acac)3 , el Fe(DBM)352c. Por otro lado, algunos autores emplean el

disolventes (ter, THF, DME y dioxano), as como distintas cantidades de alqueno,
obtuvieron el correspondiente estireno con rendimientos del 68% (va B, esquema
1.35).58 A diferencia de Kochi, observ que el producto de acoplamiento cruzado
se obtena a partir de mezclas equimolares del alqueno y del magnesiano. Por otro
lado, mientras alquenos con geometra E daban el correspondiente producto de
acoplamiento cruzado con retencin de la configuracin, los ismeros Z daban
mezclas. Aunque no se postul ninguna hiptesis, s que se observ que cuanto
mayor era el impedimento estrico del aril magnesiano empleado en las reacciones
de acoplamiento cruzado con el ismero Z, mayor era la proporcin de ismeros
del producto de acoplamiento cruzado.

alquilos de partida, la reactividad de los correspondientes cloruros, ioduros o
tosilatos de alquilo era inferior a la del correspondiente derivado bromado. Por otra

que para que la reaccin tuviera lugar en el carbono sp2 era necesario el empleo de
las condiciones de reaccin establecidas por Frstner (THF, NMP)43 si no la
reaccin no tena lugar (esquema 1.38).

Tambin observaron que los haluros primarios eran menos reactivos que los
secundarios y que los haluros terciarios daban mezclas de productos de eliminacin
y reduccin. La prdida de quiralidad observada al emplear un bromuro de alquilo
quiral aportaba indicios sobre el mecanismo de reaccin indicando la formacin de
un intermedio radicalario.
Sin embargo, habra que esperar un ao ms para que Nakamura utilizase los
reactivos de cinc como nuclefilos en las reacciones de acoplamiento cruzado de
haluros de alquilo catalizadas por hierro (esquema 1.40).63 El objetivo era superar
la limitacin impuesta por los reactivos de Grignard, es decir, su incompatibilidad

Una vez superados los problemas iniciales encontraron que la reaccin funcionaba
con derivados bromados y iodados (los clorados reaccionaban peor), el sistema era
compatible con numerosos grupos funcionales (alquenil, TMS, alquinil, steres y
nitrilos). No obstante, al igual que ocurra con los magnesianos en los alquilos
quirales se observ la prdida de la quiralidad.
En 2007 Cahiez y col. intentaron mejorar los resultados descritos hasta el momento

Mientras que la reaccin con bromuros y ioduros de alquilo secundarios tena lugar
con rendimientos elevados, el correspondiente derivado clorado no reaccionaba.
Por otro lado, este sistema cataltico permiti que la reaccin de haluros de alquilo
primarios tuviese lugar con rendimientos elevados (70%).65
Bedford utiliz tanto ligandos fosfina, fosfito, arsina como carbenos para la especie
de hierro(III) en reacciones de acoplamiento cruzado con haluros de alquilo
primarios y secundarios con hidrgenos en posicin .66 Aunque se abra la puerta
a un nuevo tipo de ligandos, los resultados obtenidos no diferan mucho de los
obtenidos con TMEDA o con complejos de hierro en bajo estado de oxidacin,
llegando a ser en algunos casos inferiores (el magnesiano con dos sustituyentes
orto en el anillo aromtico no reaccionaban).

fosfina bidentados, como la XanthPhos. Destacar que la XanthPhos es una fosfina
que presenta unas caractersticas especiales:
a) Es la nica fosfina bidentada que puede formar complejos trans, lo que
facilita la isomerizacin del complejo de Pd cis (procedente de la etapa de
adicin oxidante y requerido para que tenga lugar la eliminacin reductora) a
trans (ms estable).

Los ligandos de fosfina son los ms eficaces para estabilizar las especies de
paladio(0), siendo muy comn el Pd(PPh3)4. Sin embargo, estos sistemas presentan
una serie de problemas asociados a la incompatibilidad de las fosfinas con el aire y
el medio acuoso as como problemas asociados a los ligandos82, es decir, las
reacciones secundarias de intercambio arilo-arilo y la formacin de sales de
fosfonio. Como alternativa, se plante el uso de otros sistemas catalticos con el fin
de determinar la reactividad de este sulfxido con otros catalizadores.

Por otro lado, las reacciones de acoplamiento cruzado catalizadas por un metal en
ausencia de ligando y la catlisis en fase heterognea estn adquiriendo cada vez
ms importancia, debido a la eliminacin de las reacciones secundarias originadas
por los ligandos fosfina y la posibilidad de reutilizar el catalizador,
respectivamente.
La primera diferencia que encontramos en todos los casos con respecto al uso de la
fosfina es la necesidad de tiempos de reaccin ms largos y de temperaturas
mayores (tabla 1.2). Los resultados que se obtuvieron en la reaccin entre el 1-
bromovinil sulfxido y los cidos bornicos 2a, 2b y 2c fueron satisfactorios
(entradas 1, 2 y 3, tabla 1.2). No obstante, al emplear el cido bornico 2d el
rendimiento de la reaccin disminuye hasta el 23% (entrada 4, tabla 1.2).
Por otro lado, se emplearon fuentes de paladio cuyos ligandos fuesen capaces de
estabilizar el metal como Pd2(dba)3 y Pd(OAc)2 (entradas 5, 6 y 7, tabla 1.2). Sin
embargo, la reaccin principal fue el producto de homoacoplamiento, obtenindose
slo un 35% del vinil sulfxido -arilado o bien el producto de partida junto con
productos de descomposicin.

Por otro lado, se pens en la posibilidad de extrapolar las condiciones descritas por
Felpin para -arilacin de yodoarenos al estudio de la reactividad87 del bromo vinil
sulfxido 1a. No obstante, la reaccin de acoplamiento cruzado no tuvo lugar
cuando una mezcla de sulfxido 1a, cido p-metil fenil bornico, Pd/C, Na2CO3 en
una mezcla DMF:H2O se calent a 25C durante 24h; el producto de partida 1a se
recupera inalterado (entrada 10, tabla 1.2).

Sealar que los alquenil sulfxidos obtenidos son compuestos muy interesantes,
puesto que al escoger el cido aril bornico sustituido adecuado nos permite la
posibilidad de llevar a cabo reacciones de Suzuki-Miyaura secuenciales y acceder

Transcurridas 3h se hidrolizaron las muestras con una disolucin HCl 0.1M con el
fin de comprobar de modo comparativo la velocidad de deshalogenacin de ambos
sustratos. En la muestra correspondiente a 1c, se observa la formacin del producto
de deshalogenacin 9 (esquema 1.71), con una conversin del 100%. Ahora bien,
el resultado fue bien distinto en la muestra correspondiente a 1d, en el que se
recupera el correspondiente de producto de partida 1d inalterado (esquema 1.71).

Por otro lado, al comprobar si se produca reaccin entre la -bromo metil fenil
sulfona 1c y el bromuro de p-metoxi fenil magnesio 3a en ausencia del catalizador
se obtuvo el producto de deshalogenacin 9 en un 72%, as como el
correspondiente producto de homoacoplamiento del reactivo de Grignard 7a. Este
resultado nos sugiri la posibilidad de que un primer paso de la reaccin fuese un
intercambio metal-halgeno entre el magnesiano (I, esquema 1.72) y la -halo
sulfona en lugar de la adicin oxidante de la -halo sulfona al complejo de hierro
en bajo estado de oxidacin (II, esquema 1.72).

Con el fin de obtener evidencia experimental sobre si la formacin del producto de
deshalogenacin tena lugar mediante un intercambio metal-halgeno, se hizo
reaccionar una mezcla de la -cloro sulfona 1d con el magnesiano 3c y TMEDA a
-78C tanto en ausencia como en presencia de catalizador y se mantuvo en un bao
de hielo durante 1h. 15min. Transcurrido ese tiempo las mezclas de reaccin se
hidrolizaron con agua deuterada y se analizaron por 1H-RMN. En presencia de
catalizador, se obtuvo el producto de acoplamiento cruzado 5c deuterado, y se
recuper parte del producto de partida deuterado (I, esquema 1.73). Por el
contrario, en ausencia de catalizador se recuper el producto de partida 1d
deuterado, pero no deshalogenado.

Con todo ello se llev a cabo el siguiente ensayo; a una mezcla de -cloro metil
fenil sulfona 1d (0.5eq), 4-bromotolueno (0.5eq), TMEDA (1.2eq), Fe(acac)3
(5mol%) a -78C en THF (6mL) se aadi una disolucin de bromuro de fenil
magnesio 3b (2eq, 1M en THF) y se agit a 0C durante 3h. Transcurrido ese
tiempo la mezcla se hidroliza con HCl 0.1M. Sealar que el estudio de los
productos de reaccin se lleva a cabo por resonancia magntica nuclear de protn y
por gases-masas.

En conclusin, a partir de todos los resultados obtenidos con estos ltimos
experimentos podemos decir que la reaccin de acoplamiento cruzado no
transcurre a travs de un intercambio metal-halgeno previo entre la -halo sulfona
y el aril magnesiano. No obstante, este intercambio metal-halgeno s que tiene
lugar, siendo la entrada para la formacin del producto secundario de
deshalogenacin 9.

Por otro lado, cuando se emplean -halo sulfxidos las reacciones de acoplamiento
cruzado catalizadas por paladio dan el correspondiente producto de acoplamiento
cruzado con mayor rendimiento que las reacciones a catalizadas por hierro
(entradas 4-8, tabla 1.6). Mientras que en la reaccin del -bromo metil fenil
sulfxido y 2c se obtiene el correspondiente producto de acoplamiento cruzado en
un 81% (entrada 4, tabla 1.6); cuando la reaccin est catalizada por hierro la
quimioselectividad empeora. Puesto que, se obtienen tres productos: el producto de
acoplamiento cruzado 4o en un 31% de rendimiento, 26% del producto de ataque
directo del magnesiano al sulfxido 12 y un 41% del producto de deshalogenacin
9c (entrada 5, tabla 1.6). En este caso, el producto 12 tiene lugar a travs de una
reaccin no catalizada, es decir, ataque directo del magnesiano al azufre del
sulfxido, seguido de desplazamiento de bromometilo. Aqu, la catlisis nos
permite conducir la reaccin por una va alternativa.

Esto junto con los resultados obtenidos antes, nos confirmara que la reaccin ha
tenido lugar. Ahora bien, los productos de acoplamiento cruzado formados, podran
seguir reaccionando por el doble enlace, para generar diversos productos de
reaccin.
Por otro lado, son numerosos los estudios de sntesis de productos naturales en los
que se emplean derivados de estao como catalizador y iniciadores radicalarios
para promover adiciones radicalarias al doble enlace de la vinil sulfona.113 Aunque
nuestro sistema cataltico es hierro y no estao, ya se ha comentado en repetidas
ocasiones que, el hierro puede formar radicales. Por tanto, no sera extrao que
tuviesen lugar reacciones de adicin radicalarias de arilos al doble enlace del
producto de partida y/o del producto de acoplamiento cruzado.
Por otro lado, Vogel demostr que dependiendo de las condiciones de reaccin
pueden tener lugar reacciones de acoplamiento cruzado desulfonilativas catalizadas
por hierro entre cloruro de sulfonilo y reactivos de Grignard (esquema 1.82).114

Procedimiento general.
Sobre una disolucin de fenil vinil sulfuro (1eq, 10mmol) en diclorometano (8mL)
enfriada a -78C, se adicion bromo (1q, 10mmol) gota a gota durante 30 minutos.
Finalizada la adicin, se dej que la mezcla de reaccin alcanzase temperatura
ambiente y se adicionaron 4mL de etanol gota a gota. Se mantuvo en agitacin
durante 30 minutos, seguido de la adicin gota a gota de una solucin de KOH (1.4
eq, 14mmol) en etanol absoluto (8mL) y se mantuvo en agitacin durante 2h.
El extracto orgnico se lav la fase orgnica con salmuera (5mL); se sec sobre
MgSO4; se filtr y concentr a presin reducida hasta sequedad. El crudo de
reaccin se purific por cromatografa flash [hexano 100%] en slica gel.

Sin embargo, el oro es mucho ms barato que otros metales, como el rodio, paladio
o platino, utilizados en los procesos de catlisis a gran escala. En contraste con
estos ltimos, destacar que se reciclan miles de toneladas de oro procedentes de
aplicaciones tcnicas estequiomtricas, as como miles de toneladas son producidas
cada ao en las minas de oro.
Adems, algunos estudios demuestran que el estado de oxidacin del oro tiene una
gran influencia en la regioselectividad de la reaccin.2 Echavarren observ que la
regioselectividad de la reaccin de hidroarilacin de alquinos es opuesta en funcin
de la presencia de Au(I) o de Au(III). Georvayan observ en la sntesis de
halofuranos, a partir de reacciones de ciclacin de alenonas, la obtencin de
productos de reaccin diferentes dependiendo del estado de oxidacin del oro.3

Los estudios de RMN de 31P (CD2Cl2, 120MHz, rt) llevados a cabo en una muestra
de AuCl(PPh3)/AgSbF6 (1:1.5:1:1), 1a y 2a demuestran la existencia un equilibrio
entre las especies [AuP(Ph3)n] (singulete a 44.5ppm)82 y el complejo de oro
coordinado tanto al sulfxido como al alquino (singulete ancho a 35.9ppm).
Adems, las seales de 1H RMN correspondientes a las especies libres 1a y 2a no
se observan.

Las reacciones de acoplamiento cruzado de Stille son, junto con las reacciones de
acoplamiento cruzado de Suzuki-Miyaura, uno de los mtodos ms generales y
extendidos para la formacin de enlaces C-C. La reaccin de Stille utiliza
organometlicos de estao y haluros de arilo/alquilo y en contraste con las
reacciones de Suzuki no requiere condiciones bsicas pudindose llevar a cabo la

En consecuencia, el siguiente experimento se realiz en presencia de una cantidad
cataltica de paladio y cobre (esquema 3.32). As, una mezcla del ter de oxima 1a,
fenil acetileno 2a, Pd(OAc2)3, XantPhos y CuI en NEt3 se calent a reflujo durante
16h. Se obtuvo el diino procedente del homoacoplamiento del alquino y el
producto de partida.

Este resultado coincide con los obtenidos por Buchwald en las reacciones de
alquinilacin de cloruros benclicos34. Este autor observ que la adicin de CuI en
cantidades catalticas inhiba la reaccin de alquinilacin deseada, vindose
favorecida la formacin de productos secundarios.

Por otro lado, de acuerdo con los resultados obtenidos en las reacciones de
hidratacin de los teres de oxima catalizadas por oro y por cido, como primera
hiptesis proponemos que el grupo oxima es la responsable de la regioisomera
observada y la total selectividad con la que transcurre la reaccin. Para tratar de
evidenciar esta hiptesis se plane contrastar las oximas proparglicas con las
reacciones de hidratacin de 2-alquinil metoxibencenos, para observar si la
presencia del grupo metoxi tiene el mismo efecto que el grupo C=NOMe en la
orientacin de la adicin del agua.

De acuerdo con estos datos una posible en una primera hiptesis proponemos que
el ter de propargil oxima 3 en presencia del catalizador (oro o cido p-
toluensulfnico) se isomeriza a 3 (esquema 3.46), siendo sta la especie que se
hidrata.

Por otro lado, en las reacciones de hidratacin de los teres de oxima proparglicos
3l y 3m (grupo t-Bu en el ter de oxima) en los que la isomerizacin no puede
tener lugar por motivos estricos, se recuperan los productos de partida. Lo cual
apoyara la hiptesis de que la reaccin de hidratacin nicamente tiene lugar
cuando se asocia a la isomerizacin (Z)-(E).
307 Resultados y discusin
El ciclo cataltico propuesto para la reaccin de hidratacin catalizada por oro (III)
consta de 5 etapas (esquema 3.47). As pues, el mecanismo propuesto supone una
primera etapa de coordinacin del oro al nitrgeno del ter de la oxima generando
el intermedio de reaccin I, seguido de la isomerizacin del (Z)-ter de oxima a
(E)-ter de oxima y coordinacin del catalizador al enlace triple formando el
intermedio II. Tras el ataque del metanol al enlace triple y evolucin al intermedio
III, hay un segundo ataque nucleoflico, bien de otra molcula de metanol bien de
una molcula de agua (IV). Finalmente, tras la hidrlisis y protodemetalacin del
intermedio IV se obtiene la cetona 8 que isomeriza a 8. Asimismo, tras la etapa de
protodemetacin se regenera el catalizador y puede reiniciarse un nuevo ciclo
cataltico.

Por su parte, en las reacciones de hidratacin catalizadas por cido p-
toluensulfnico el mecanismo de reaccin constara de las etapas de coordinacin
del protn al nitrgeno, isomerizacin(E)-(Z) del ter de oxima, coordinacin del
catalizador al triple enlace y ataque del metanol o agua (esquema 3.48). A
diferencia de la catlisis con oro, aqu tras la formacin del intermedio VI se
liberara un protn que coordinara a otro ter de oxima.

A wide variety of methods are available for the synthesis of ethers, and among
them points out the Williamson ether synthesis because for years has been
considered as the best general method for the preparation of symmetrical and
unsymmetrical ethers. Alternatively, ethers are also prepared from alcohols using
Brnsted acid catalysts.5 The reactions using Brnsted acid are often conducted
under harsh conditions that lead to the mixture of products. Moreover these are
predominantly focused on the synthesis of symmetrical alkyl ethers. On the other
hand, different catalytic systems based on transition metals, which can be
considered as promising methods, are reported for etherification of alcohols even
though only a reduced number of metals have been investigated to date. In general,
the Lewis acid reagents used for the preparation of ethers are in homogeneous

phase and some of them endure from reusability. However, some of the Lewis acid
catalysts are deactivated by water, which is produced during the reaction. Thus, in
order to overcome the difficulties associated with homogeneous catalysts, usage of
heterogeneous catalysts, such as resins6, zeolites and heteropoliacids7 has been
reported.
In addition, ethers can be obtained from the reaction of an alcohol and carbonyl
compounds,8 the reaction of a silyl ether and an aldehyde9 and even through a
isomerization of oxo-alkynes.10
However, in this chapter are only detailed the methods which use two alcohol
molecules to form an ether.
Thus despite the current methods, the development of new catalysts which are
readily available, active at low loadings and which offer a broad scope and
selectivity, is still an open objective in the synthesis of ethers from alcohols.

Preparation of ethers by this methodology was most successful when the alkyl
halide was reactive toward SN2 substitution. Methyl halides and primary alkyl
halides were the best substrates. Secondary and tertiary halides were not suitable,
because they tended to react with alkoxide bases by E2 elimination rather than SN2
substitution. Unsymmetrical ethers should therefore be synthesized by reaction
between the more hindered alkoxide partner and the less hindered halide partner
rather than vice versa, being a clearly dependence on the nature of the alkyl halide.
However, this reaction most times is not applied industrially due the fact that the
starting halide compounds are not easily available or they are too expensive.
Furthermore, it should be also considered that the use of the starting halides in
addition to the stoichiometric amounts of waste salts, becomes this method in the
less environmentally desirable process to provide ethers.
A useful variation involves silver oxide or silver carbonate, as base rather than
sodium hydride. Under these conditions, the alcohol reacted directly with the alkyl
halide, so there was no need to perform the metal alkoxide intermediate. For
example, glucose reacted with iodomethane in the presence of silver Ag2O to
generate a pentaether (equation 4.2).

It was also possible to mix the halide and the alcohol, pyrrol, indol or phenol
directly with Cs2CO3 in acetonitrile12 or with solid KOH in Me2SO13. On the other
hand, in order to use less expensive reagents the reaction could take place in the
presence of sub-stoichiometric amounts of HgO/TBFA in CH2Cl2 in high yields.14
Treatment of tertiary alkyl halides with zinc acetate and ultrasound leaded to the
corresponding acetate in a related reaction.15 They could be also prepared from
alkyl halides by treatment with diethylenglico ditosylate (instead of phenol) in
presence of K2CO3 and a crown ether (equation 4.3).16

In addition, other alkoxide sources, such as acetates17 and mesylates18 have been
used in the presence of catalytic amounts of metals, to give the corresponding
ethers.
One of the first methods employed for the synthesis of ethers, and which is
traditionally described in organic chemistry books19 is the dehydration of alcohols

When a primary alcohol was heated with sulfuric acid, the symmetric alcohol was
produced in high yield. Although the detailed mechanism was unknown, the
reaction could take place by two possible pathways. One possibility was a reaction
between the alkyloxonium ion, formed by reaction with the sulfuric acid, and
another molecule of alcohol (via A, equation 4.4). Alternatively, the alkylsulfuric
acid formatted could react with another alcohol to give the corresponding ether (via
B, equation 4.4).

On the other hand, when this methodology was extrapolated to secondary or even
tertiary alcohols the elimination product was obtained as the majority one.
However, in 1932 Norris20 reported the synthesis of unsymmetric secondary and
tertiary alkyl ethers catalyzed by sulfuric acid. The better yield was obtained when
t-butanol (1mol) was added to a boiling mixture of ethanol (2mol) and sulfuric acid
(15% aqueous solution). In contrast to this, isobutylene was obtained in large
quantities due the minor reactivity of secondary alcohols. In order to improve these
results, higher quantities of the secondary alcohol were required, as well as sodium

A few years later, Smith21disclosed the etherification of carbinol with primary and
secondary alkyl alcohols (methanol, isopropanol, propanol) in high yields (86-
97%). But, more complex alcohols such as t-butyl and benzyl alcohols did not react
probably due the effect of the branched-chains of the alcohol and a higher stability
of the carbinol than the corresponding ether in the reaction mixture.

On the other hand, Ito et al.23 reported the reaction between aliphatic alcohols and a
phosphonium salt (figure 4.1) to give the corresponding symmetric ether. Primary
alkyls gave ethers in moderated yields (11-60%), while secondary and tertiary
alcohols due their steric hindrance gave exclusively olefins, also in poor yields.

In spite of the fact that, Mukaiyama et al. disclosed some years earlier a oxidation-
reduction condensation methodology, this was not applied to the synthesis of ethers
until 2003.24 Its fundamental concept was to perform dehydration condensation by
removing H2O in elements of 2[H] and [O] with the use of a weak reductant-
oxidant combination.25 The purpose of the oxidant was to convert the phosphonium
salt to the pentavalent phosphorous intermediate (I, equation 4.6 and III, equation
4.7) by catching one hydrogen atom from alcohols or acids and evolve to give the
corresponding ether or carboxylate. In addition, there are two main differences
between this methodology and the one described above, the first one is that the
reaction was carried out under mild and neutral conditions in absence of any
assistance of acids or bases and the second one was that no racemization occured.

unexplored. However, it has been pointed out that low and high oxidation state of
rhenium-oxo complexes have been successfully used in etherification reactions.

The mechanism proposed (Scheme 4.1) was based on the MTO oxophility,
established in precedent studies related to MTO-catalyzed oxidation of alkynes29,
cobalt thiolates30, organic sulfides31, anilines32, alkynes33 and phosphines34.

The lack of either elimination products or the symmetric ether obtention implied
the formation of a benzyl cation, which would be the crucial step in the formation
of the unsymmetrical ethers. Moreover, when the reaction was carried out under an
inert atmosphere, the corresponding ethers were obtained in low yields (<5%),
indicating a previous oxidation of the catalyst by oxygen to give a rhenium(III)
oxide.
On the other hand, Toste36 has disclosed the etherification of propargyl alcohols
using a rhenium(V) oxo-complex bearing a bidentate phosphine ligand as the
catalyst and acetonitrile as solvent. Substitution occurred with a wide variety of

However, a few years later Sharma39 demonstrated that FeCl3 (10mol%) in CH2Cl2
at room temperature is an efficient reagent for the conversion of primary and
secondary aliphatic alcohols, acid sensitive ethers and ketals into diphenylmethyl
ethers (DPM).
I.2.3.3.- Rare earth(III) catalyst system.
Rare earth(III) salts such as triflates40 have been advantageously utilized as
versatile Lewis acids in several organic reactions. They differ from conventional
Lewis acids, due their stability in water and their no decomposition under aqueous
work-up conditions. Moreover, it has been demonstrated that the catalyst can be
easily recovered and reused without decrease in its catalytic activity.41
In 1999, Sharma42 reported the protection reaction of an alcohol (1eq.) with p-
methoxybenzyl alcohol (2eq.), catalyzed by Yb(OTf)3 (5mol%) in CH2Cl2 at room
temperature, obtaining the corresponding ether in high yields, and 10-15% of the p-
methoxybenzyl symmetrical ether. This protocol disclosed the compatibility to
acid, and alkali-labile protecting groups and their tolerance of a wide variety of
functional groups (THP, TBS ethers and esters) and substrates, such as primary and
secondary aliphatic alcohols, acetonides and terpenes. Although, no reaction was
observed when tertiary alcohols were used (equation 4.10).

A few years later, Sharma39extended the scope of the reaction to p-methoxy
diphenyl methanol (MDPM), p-phenyl diphenylmethanol (PDPM) and p-phenyl
diphenylmethanol (PPDPM) as protecting groups for alcohols by reducing the
quantity of the benzylic alcohol to 1 equivalent and raising the catalyst to 10
mol%.41 Furthermore, this secondary and tertiary benzylic alcohols reacted with
sugar alcohols, terpenes, 1-2 and 1-3 diols, in these last cases giving the mono an
di-protected ethers.
Parallel, several studies with other lanthanide(III) triflates as catalysts have been
carried out. In 2002, Kawada43 demonstrated that the etherification reaction
between hydroxybenzyl alcohols (1eq) and primary, secondary and even tertiary
alcohols (5eq) in toluene at 80C could be catalyzed by many rare earth metals (Sc,
Y, La, Pr, Nd, Sm, Eu, Gd, Dy, Ho, Er, Tm, Yb, Lu) in high yields. A deeper study
revealed that Sc(OTf)3 was a more active catalyst than Yb(OTf)3, decreasing the
first one its catalytic activity with the extension of the reaction time. However the
reaction did not proceed when o-substituted benzylic alcohols were used, because
of the stabilization of the reactant by bidentate chelation of Yb(OTf)3 (equation
4.11).

sources of silver were tested, finding that AgPF6 afforded much lower activities,
probably due to the generation of dicationic iridium species which provide a lower
electron density than the AgOTf. Only for the homocoupling of benzyl alcohol was
obtained a low yield, due to the favorable formation of the benzaldehyde. It is
appropriate to underline that in all cases the corresponding symmetrical ethers were
obtained under 16% yield (equation 4.13).

On the other hand, the mechanistic pathway proposed occurs through a typical
organometallic catalytic cycle mixed with an acid-catalyzed process in which the
cationic H-IrV intermediate acts as a protic acid.
In the literature has been reported that benzylic, allylic and tertiary alcohols
can be activated with zinc to generate carbocationic species, which are trapped
with different nucleophiles.46,47,48,49 This prompted Kim50 to study the etherification

catalyzed by Pt(II) salts proceeded through an oxidative addition of the methanol

A few years later, Pale54 obtained similar results when a PdCl2 source was used
either in solvent free conditions or in the presence of non-coordinating solvents.
Nevertheless, they found that acetronitrile allowed the reaction while being a polar
solvent, due its ability to coordinate and stabilize Pd species.
On the other hand, Ikariya55 disclosed the symmetric and non symmetric
etherification reaction of allylic alcohols and primary or secondary alkyl alcohols
catalyzed by Pd2(dba)3/P(OC6H5)3 at 80C in chloroform. In contrast to these
results, allylation of bulkier tertiary alcohols gave unsatisfactory results (equation
4.16).

Nevertheless, when the reactions were carried out with chiral alcohols, a mixture of
two diasteroimeric isomers was obtained in high yields. Further studies confirmed
that the propargylic substitution reactions proceeded via allenylidene complexes
which had been characterized by X-Ray (figure 4.2).

In their first studies observed that a medium-sized cyclic ethers could be thought
the intramolecular attack of a hydroxyl group located in a saturated chain to a
carbocation generated by acid treatmen of propargyl-cobalt complexes (I, figure
4.3).59a A high degree of stereocontrol was reached when defined stereocenters
were present and was obtained only one of the possible diasteroisomers.

On the other hand, this reaction was extended to the intermolecular synthesis of
propargyl ethers59b (II, figure 4.3). In order to minimize the formation of the dimer
III (figure 4.3), resulting from the attack of the propargylic alcohol itself to the
generated cation, high dilution condition and an excess of the nucleophilic alcohol
must be used. This method proved also to be a general way to obtain mixed ethers
when primary alcohols were used. On the contrary, the use of secondary or tertiary
nucleophilic alcohols a very low yield was obtained, probably due to competitive
elimination reactions. Moreover, Martin59c spread out the use of dicationic species
to the synthesis of linear and cyclic dipropargyl ethers (IV, V, figure 4.3).
In 2000, Boyer 60 disclosed that BiBr3 was efficient in promoting the benzylation of
aliphatic alcohols (primary, secondary and tertiary) by various benzylic alcohols
such as benzyl alcohol, 1-phenyl ethanol and 2-phenylpropan-2-ol in CCl4 at room
temperature.
As seen in figure 4.4, the mechanism for benzylation would involve the activation
of its hydroxyl function, through a complex with BiBr3, followed by the hydroxylic
attack of the hydroxy group of the benzylic alcohol bonded to the same bismuth
atom. Furthermore, the racemization could be explained as consequence of two
possible attacks of the nucleophilic alcohol. Attack from side a involving inversion
on the aliphatic alcohol and attack from side b implying retention.

To explain the reaction pathway, Gillis suggested a cyclic transition state, in which
DMSO aids in the removal of a proton from the attacking group while
simultaneously stabilized the leaving group (I, equation 4.20).
In 1977, Emert66 expanded the scope of the reaction to the synthesis of symmetrical
benzyl ethers in high yields. In contrast with the results obtained when ZnCl267 was
used as catalyst, dehydration of p-methoxybenzyl alcohol gave the corresponding
ether instead of the polymerization products observed before. Nevertheless, this
method possessed a limitation because no product was obtained upon heating p-
nitrobenzyl alcohol in DMSO. Thus, the radical decomposition of DMSO in strong
acids would quench the nitro group, inhibiting the reaction.

Solid supports have found wide applications in organic reactions, because of the
easier work-up of the reaction mixtures and usually high selectivity accompanied

most part of the previous proposals. This mechanism allowed a common
intermediate to produce either an ether or an alkene, an alcohol could absorb to act
as nucleophile or electrophile, explained the stereochemistry obtained and allowed
the obtention of mixed ethers (scheme 4.2).

On the other hand, alumina has also been used as a support for transition metal
oxide catalysts such as molybdenum, chromium and vanadium being the main
metal-support interaction through the hydroxyl groups surface of the support.
However, when this methodology was applied to the synthesis of dibenzyl ether74 a
different catalytic activity was observed, depending on the supported metal, giving
molybdenum supported catalysts the highest yield (82%).
that selectivity directed to the synthesis of ethers was also strongly dependent on
the length of the alcohol chain and on temperature, those were favored at low
temperatures while alkenes were obtained at high temperatures.
A few years later Ballantine76 studied the liquid phase dehydration of alcohols over
montmorillonite catalysts at 180C, and concluded that primary alcohols underwent
preferential intermolecular dehydration to give the corresponding dialkyl ethers in
high yields while secondary alcohols favored the formation of the alkene via an
intramolecular dehydration pathway. However, benzylic alcohols produced poly
(phenylene methylene) in presence of ion-exchanged montmorillonites.

On the other hand, modified zeolites have been widely used for the synthesis of
fuel additives such as ethyl t-butyl ether79, dimethyl ethyl ether80 or t-amyl
alcohol81 in high yield. However, undesirable side products such as coke are
formed due to the presence of strong acid sites and the high dehydration
temperature. Thus, research has been focused on finding better catalysts that give
higher selectivity towards the ether.
In 1993 Klier disclosed the synthesis of methyl isobutyl ether from isobutanol and
methanol over Nafion-H, Amberlist H, H-mordenite, H-ZSM-582 and (HO)3Zr-

O3SCH2CH2OSO3-Zr(OH)3 at high pressure. Further kinetic studies related to
isotopic labeling and chiral inversion experiments indicated that the reaction was
dominated by the surface SN2 reaction pathways through the oxonium ion
intermediate. 84
A few years later, Olah85 reported the use of Nafion-H as a highly efficient solid
acid catalyst for the bimolecular conversion of alcohols to symmetric ethers.
Primary aliphatic alcohols gave the corresponding ethers in high yields, while
secondary cyclic and acyclic alcohols gave a mixture of structural isomers. The
mechanism for the reaction involved the in situ formation of an oxonium ion,
where the Nafion-H converted the hydroxyl group of the alcohol in a better

A few years later, Tarlani et al.88 disclosed the synthesis of primary and secondary
benzyl ethers catalyzed by H6P2W18O62 in dichloroethane at 80C (34-90% yield).
It must be enhanced that only secondary benzyl ethers which were not able to give
elimination products succeeded in the etherification.
On the other hand, associated with HPAs are some disadvantages like low thermal
stability, low surface area and solubility in polar solvents. Thus, in order to solve
this, the strength of the HPAs could be tuned by exchanging the protons by metal
cations. This concept was employed by Firouzabadi et al.89 and Lingaiah90 in the
etherification of benzylic alcohols (primary, secondary and even tertiary) with
aliphatic and benzylic alcohols by aluminumdodecatangstophosphate (AlPW12O40)
or cesium exchanged tungstophosphoric acid supported on zirconia, respectively.

Thus, attempting to this precedents and in order to extend the scope of the gold-
assisted addition of oxygenated nucleophiles to triple bonds, we decided to study
the addition of benzylic alcohols to terminal alkynes to obtain dihydrobenzofuranes
(equation 4.24).

Our aim consisted of taking advantage of the positive charge generated by the
addition of gold to the multiple bond. As it is described in the literature, a firstly
addition of the gold to the terminal carbon of the alkyne would generate a positive
charge, and as consequence this electrophilic carbon would suffer a nucleophilic
attack of the oxygen of the alcohol (I, scheme 4.4). Once here, what we expected
was a second coordination of the alcohol to the double bond (II, scheme 4.4),
followed by an intramolecular nucleophilic attack of the carbon, generating a
dihydrobenzofuran (III, scheme 4.4).

In our first essay we tested the addition of benzyl alcohol 2a (50 eq., 20 mmol.) to
phenyl acetylene (1eq, 0.4 mmol) catalyzed by NaAuCl4 (5 mol%) at 70C during
3h, but surprisingly the reaction did not undergo to the synthesis of
dihydrobenzofuranes. Moreover, the starting alkyne was recovered and the only
reaction product obtained was the symmetric ether of benzyl alcohol (equation
4.15).

Similar results were obtained when indanol was used. However, the use of benzyl
alcohols with electron withdrawing groups, such as, p-bromo did not give the
symmetric ether, starting materials were recovered unchanged.

Despite the fact that the obtained results are far away from the previously proposed
objective, the obtention of the symmetric ether encouraged us to consider the scope
of the synthesis of unsymmetric ethers, due their relevance in the perfumery
industry and the fact that today scientifics keep on the search for simple and
efficient ether synthesis
The results obtained in the addition of alcohols to alkynes suggested us that the
reaction was based in a carbocationic intermediate.95 Hence, the preparation of
unsymmetrical ethers requires an appropriate catalytic system that is capable of
distinguishing between two identical functions and developing a different role for
each partner. This condition is an essential requisite to control the selectivity in the
reaction, otherwise a mixture of the three possible products (unsymmetrical ether,
symmetrical ethers R-O-R, R-O-R) is potentially expected. As consequence, the
stronger contrast in the components of the alcohols the greater possibility the
catalyst has of choosing one of them to play a differentiated role (equation 4.26).

So as to obtain the desired unsymmetrical ethers, we have split the starting alcohols
in two groups: the alcohols which are able to generate a stable carbocation, such as
benzyl alcohols or tertiary aliphatic alcohols, and the alcohols which would act as
nucleophiles, thats primary aliphatic alcohols.

In order to start the runs benzylic alcohols were chosen, due their importance in
chemistry like protector groups and the potential stability of their carbocations.
First of all primary benzylic alcohols were tested and later on, more complex
secondary alcohols, such as indanol and the precursors of protecting groups (9-
fluorenol, benzydrol, methyl DL-mandelate, sec-phenetyl alcohol.
Initially, we checked the reaction of the highly reactive primary p-methoxy benzyl
alcohol 2a with the less reactive aliphatic primary, secondary and tertiary alcohols,
which are used in a higher concentration than the benzyl alcohol to direct the
reaction toward the formation of the unsymmetrical ether.

Once the optimal conditions were determined, we evaluated the substrate scope of
this new gold catalyzed-etherification reaction. All the reactions were run only for
1-3h in order to examine the difference between the different systems employed
(equation 4.27).

The reaction of 2a with primary unbranched alcohols (4c, 4d) gave the formation
of the ethers 5ac, 5ad, respectively, with moderate yields (Table 4.2, entries 3 and
4).

On the other hand, more complex primary alcohols such as the -branched (4a)
and the -branched (4b) ones were tested too. It is interesting to notice that in these
cases the yield not only was kept (table 4.2, entries 3), moreover, it increased until
80% (table 4.2, entry 3). Although alcohols 4a and 4b can suffer transpositions,
and as consequence gave other reaction products, the only products obtained were
the unsymmetric ethers. Here, we found a marked difference with protic catalysts
used in etherification reactions, because gold shows a higher control in the side
reactions than protic acids that usually give transposition products. Though, this
methodology presents an elevated selectivity.
However, the result was different when secondary alcohols (4e) were used. The
reaction proceeded with a low yield due the steric hindrance from the alcohol,
recovering the starting benzylic alcohol.

It is worth mentioning that in our case, and in sharp contrast with the precedent
methods, the unsymmetrical ether was always the main product without any
contamination of the symmetrical ether of the benzyl alcohol. In addition, the
adequate excess of the aliphatic alcohol 4 used was sufficient to preclude the
reaction of alcohol 2a with the water that built up in the system, thus allowing the
reaction to take place without the need of dehydrating agents.
On the other hand, an improvement in the yields of the secondary alcohol 4e has
been observed (table 4.3, entry 2). And since, this alcohol cannot suffer any kind
of transposition a higher concentration of benzylic carbocation favors the reaction
with the aliphatic alcohol. Due the results obtained, we decided to try the
etherification reaction with more hindered alcohols, such as the tertiary alcohols t-
amyl alcohol and t-butanol. Owing to the steric hindrance the unsymmetrical ethers
were produced in moderate yields (table 4.3, entries 4 and 5).

catalyst took place with moderate and low yields, respectively (table 4.5, entries 1
and 2). Here the steric hindrance either of the aliphatic alcohol either of the indanol
have to be considered in order to explain the obtained yields. Moreover, as
difference with the p-methoxybenzyl alcohol indene was obtained as a side
product.

However, this side product disappears when the catalyst loading is increased until
5mol%. Moreover, the conversion of the alcohol into the ether compound was
really improved and the yields increased until more than 80% (table 4.6, entries 1
and 2). Similar results were obtained with the cyclic secondary alcohol 4i showing
that the increase of the amount of catalyst favors the etherification reaction instead
of the formation of the side product 6.

Although the first experience carried out was disappointing (table 4.8), the
obtaining of a 34% yield in the etherification of a more hindered benzylic alcohol
encouraged us to beat the reaction conditions. In this case, a simple increase of the
temperature contributed to enhance the yield until 56% (table 4.9, entry 1).
Anyway, once the optimal conditions were determined, we evaluated the substrate-
scope of this reaction. Secondary benzylic alcohols 2c-f reacted with isopropanol to
give the ethers 5ce-5fe.

Thus, the substrates 2c-f and isopropanol posses bulky groups are not surprising
the moderate obtained yields (table 4.9, entries 1, 3 and 4). The steric hindrance of
the benzylic alcohol and of the isopropanol difficult the coordination of the oxygen
atom to the gold complex.
It is interesting to notice that in the purification are only obtained the unsymmetric
ethers and the starting materials which have not reacted. However, the ether from
the reaction of methyl DL-mandelate with isopropanol could not be isolated an
dully characterized, because it decomposes during the purification. Therefore, the
given yield belongs to the 1H NMR mixture crude (table 4.9, entries 2).

The corresponding ethers were obtained with modest yields (table 4.10, entries 1
and 2). These results were not surprising because the carbocations derived from
tertiary alcohols (2h and 2g, respectively) are less stable than the benzylic ones.
That means that the reactions are more difficult. Despite the yields obtained, these
results represent the first examples of reactions between two non benzylic alcohols
catalyzed by a transition metal.

On the other hand, in order to test the scope of the reaction further, we attempted
an etherification reaction arising from a mixture of an aliphatic secondary and a
primary alcohol. Unfortunately, the catalyst was ineffective and the starting
materials were recovered unchanged, even under forced conditions (table 4.10,
entry 3).
Therefore, the tertiary alcohol represents the borderline point regarding the
capacity to generate the electrophilic partner needed to accomplish etherification
reaction.
Vanillin is a natural product which has been extensively used in flavours and in the
fragance industry. We found it an interesting compound to test, due its importance
and the presence of another sensitive functional group in the molecule (equation
4.28).

Thus, we checked the reaction between vanillin (2g) with a secondary aliphatic
alcohol (4e) catalyzed by gold(III). In spite of the expectations put in this substrate,
the unsymmetric ether was not obtained. However, the appearance of side products
derived from the nucleophilic substitution in the benzene ring were observed.

Since, the dehydration reaction between two alcohols catalyzed by gold(III) has not
been described before, we found it should be interesting to contribute some
evidence on the reaction mechanism.98

- Proposed mechanism.
The reaction starts with the coordination of the oxygen atom to the benzyl alcohol
(or tertiary alcohol) to the catalytic species, which can be either NaAuCl4 or AuCl3,
this last is obtained by the loss of a chloride atom from the salt (scheme 4.6, step
A). Next, a transfer of the hydroxyl group to the metal would generate the
intermediate carbocation (scheme 4.6, step B), followed by the trapping of the

The HCl produced in the reaction media could be the responsible of the observed
etherification reaction, in would imply that gold is not the real catalyst. To verify
which the specie catalytic is, several control experiments have been performed.

In general, much less efficient transformations were achieved when reactions were
catalyzed by HCl (table 4.11). For instance, the reaction with 1a and 2e gave a
mixture of unsymmetrical and symmetrical ether (table 4.11, entry 1). This result
contrast with the reaction catalyzed by gold(III), due a higher conversion was
observed in the corresponding symmetric ether (72% yield). Moreover, no
symmetric ether was obtained. A similar yield was obtained when indanol was
employed (table 4.11, entry 2). However, in this the elimination of hydrogen

produced indene as a side produced instead of the symmetric ether of the indanol.
On the other hand, no reaction took place between tertiary alcohol 2g and
isopropanol, recovering the starting materials (table 4.11, entry 3).
This experiments demonstrate that gold(III) is the catalytic specie not the hydrogen
chloride generated in the reaction mixture.
In recent years, efforts in preparative chemistry have focused not only on the
development of new processes, but also on improving the sustainability of existing
transformations.99 In transition metal catalysis, this involves the decrease of the
catalyst loading, recycling of the catalyst, and use of environmentally friendly
reaction media.

noteworthy to mention that this methodology was adequate for alcohols bearing
either a terminal or internal alkyne group.

On the other hand, one of the most benign solvents is water due its physical and
chemical properties and its harmless and environmentally friendly character. But it
is rarely used in transition metal catalysis since many catalysts are highly sensitive
towards moisture.
Moreover, only two examples where the etherification reaction is carried out in
aqueous media have been reported. In 2002 Kobayashi described the first
dehydrative symmetrical ether formation from two alcohols in water catalyzed by
dodecylbenzenesulfonic acid (DBSA) (equation 4.31).102

So, this background and the results described before prompted us to study the
etherification reaction in water. Nowadays, we have a few preliminary results
which are encouraging.

With this reaction conditions, we evaluated the substrate scope of the etherification
reaction in water. Either 2a or 2b gave the corresponding unsymmetric ether in
high yields (table 4.12, entries 2 and 4). Moreover, the secondary benzylic alcohol
2c gave the unsymmetric ether 5be in 51% yield (table 4.12, entry 6). The
comparison of the obtained yields in homo- and heterogeneous catalysis is also
interesting. At this point, we found that similar results were obtained in both cases
(table 4.12, entries 1-2, entries, 3-4 and entries 5-6).

A mixture of alcohol 1a, alcohol 2 and NaAuCl4 was added to a flask fitted and
was heated to 70C. After being refluxed for 1h, the reaction mixture was cooled to
room temperature, diluted with ethyl acetate (5mL), and filtered over Aluminium
oxide activated. Then the solution was evaporated under reduced pressure.
