Procedure Assay Reaction

All samples and standards should be run in duplicate. 1. Add 100 L of Cu2+ Working Solution to all standard
and sample wells.
Trolox Standards for Colorimetric Detection
Add 0, 4, 8, 12, 16, and 20 L of the 1 mM (1 nmole/L) 2. Mix well using a horizontal shaker or by pipetting,
Trolox Standard Solution into a 96 well plate, and incubate the reaction for 90 minutes at room
generating 0 (blank), 4, 8, 12, 16, and 20 nmole/well temperature. Protect the plate from light during the
standards. Add water to each well to bring the volume incubation.
to 100 L.
3. Measure the absorbance at 570 nm (A570).
Sample Preparation
Serum, urine, and culture media samples can be
directly added to the wells.

For serum samples, it is suggested to assay

0.010.1 L without Protein Mask or 110 L with
Protein Mask.

For small molecule TAC, dilute samples at 1:1 ratio with

Protein Mask.

For unknown samples, it is suggested to test several

sample dilutions to ensure the readings are within the
linear range of the standard curve.

Add 1100 L of sample into wells. Bring samples to a

final volume of 100 L with water.

Results
Calculations Concentration of Trolox Equivalents
The background for the assays is the value obtained for Sa/Sv = C
the 0 (blank) Trolox Standard. Correct for the
background by subtracting the 0 (blank) standard value where:
from all readings. Background values can be significant Sa = Trolox equivalent of unknown sample well
and must be subtracted from all readings. Use the (nmole) from standard curve
values obtained from the appropriate Trolox Standards Sv = Sample volume (L) added into the wells
to plot a standard curve. C = Concentration of antioxidant in sample (nmole/L
Note: A new standard curve must be set up each time or mM Trolox equivalents)
the assay is run.
Sample Calculation
Using the corrected measurement, Trolox equivalents Amount of antioxidant = 4.84 nmole
of the sample may be determined from the standard (from Trolox standard curve)
curve.
Sample volume (Sv) = 50 L

Concentration of antioxidant (Trolox equivalents) in

sample:

4.84 nmole/50 L = 0.0968 nmole/L
