GTB 212 PRACTICAL 1: A. Microscopic and staining of microorganism

LAY OUT PLAN FOR TASKS AND STUDENTS GROUP


The laboratory practical involves 4 main tasks:

Task 1: Part A: Hanging Drop Slide & Motility


Task 2: Part B: Simple Staining
Task 3: Part C: Differential Staining- Gram Stain and Acid Fast bacilli Stain
Task 4: Part C: Differential Staining- Capsule Staining & Fungal Staining

Students will be divided to 2 major groups (Biomedic and Nursing group).

1. Biomedic group - all in MSA 1- divide to 4 subgroups


2. Nursing group - all in MSA 2- divide to 4 subgroups.

The subgroups will rotate and go to every table in the assigned group laboratory
only and fill up the practical handout.

Makmal Sains Asas 1- Biomedic Group- 4 subgroups- Table 1, 2, 3 and 4

Makmal Sains Asas 2- Nursing Group- 4 subgroups Table 1, 2, 3 and 4

Table 1 - Task 1: Examine and record the results for hanging drop and
motility of bacteria
Three Bacteria are:
a. Vibrio cholerae
b. Candida albicans
c. Escherichia coli

Table 2- Task 2: Examine and record the results of Simple Staining for:
a. Staphylococcus aureus
b. E. coli

Table 3- Task 3: Differential Staining


a. Gram Staining: Examine and record the results of Gram Staining
for:
1. Staphylococcus aureus
2. C. albicans
3. E. coli
b. Acid Fast Bacilli Staining: Examine and record the results of AFB
Staining
1. Mycobacterium tuberculosis

Table 4- Task 4: Differential Staining


a. Capsule staining - Indian Ink Preparation for Cryptococcus
neoformans
b. Fungal Staining- Cellophane tape technique by using
Lactophenol Cotton Blue (LPCB)

Students will be watching the video on:

a. Hanging Drop technique

b. AFB Staining procedure

c. Media pouring plate technique

d. Aseptic technique of Culture transfer for plate, tube bottle media (solid,
semi solid and liquid) and subculture of microorganisms.
