What is Biotechnology?
Simply put, biotechnology is the manipulation of living
things to create a product that is useful for humans.
Biotechnology has its roots in traditional practices that
have been used since the ancient times. For example,
fermentation has been used to preserve food for over
6000 years. Scientists later discovered that microorganisms
are responsible for fermentation. Fermented products like
yogurt, sauerkraut and wine continue to be consumed on
a regular basis. Furthermore, selective breeding techniques
have been used to promote desirable qualities in livestock
and food crops like corn. Early farmers may have crossed
a corn plant with larger kernels with another plant that
produced more avorful corn (Figure 1, top). Plants in the
next generation would produce corn with large, delicious
kernels (Figure 1, bottom).

1. DILUTE 50X buffer with distilled water to create 1X buffer (see Table A).

3. DISSOLVE the solution by boiling in the MICROWAVE for 1 minute. REMOVE ask and MIX by swirling.
Continue HEATING in 15-second bursts until completely dissolved and solution is clear.
4. COOL to 60 C with careful swirling.
5. SEAL the gel-casting tray with rubber end caps. PLACE the comb in the appropriate notch.
6. POUR cooled agarose into prepared gel-casting tray. WAIT 20 minutes for the gel to solidify.

Work?
This simulation experiment demonstrates the process of DNA amplication
by PCR and how the amplied product is detected by separating the reaction
mixture by agarose gel electrophoresis.
It Work?
Quantitative real time PCR (qPCR) is used to determine the amount of a spe-
cic target DNA at the same time they are amplifying it. In this simulation,
students will explore the principles of qPCR using colorful dye samples. Using
agarose gel electrophoresis, they observe the relationship between cycle num-
ber and the quantity of DNA present within the sample. Students will perform
data analysis to support these observations, making it easy to incorporate STEM
into your classroom. No qPCR instrument is necessary!

DNA sizing is an excellent tool used in many biotech applications, such as DNA

Plasmid DNA
Small-scale rapid isolation of plasmid DNA is a routine procedure used for
screening and analysis of recombinant DNAs in cloning and subcloning
experiments. In this experiment, students isolate plasmid DNA without the
use of toxic chemicals such as phenol or chloroform.

To perform PCR, the template is mixed with primers, Taq polymerase and nucleotides. The mixture is heated to 94C
to denature the DNA duplex (i.e., unzip it into single strands). Next, the sample is then cooled to 45 C - 60 C, al-
lowing the primers to base pair with the target DNA sequence (called annealing). Lastly, the temperature is raised
to 72 C, the optimal temperature at which Taq polymerase will extend the primer to synthesize a new strand of
DNA. Each PCR cycle (denaturation, annealing, extension) doubles the amount of the target sequence in less than
ve minutes. In order to produce enough DNA for analysis, twenty to forty cycles may be required.

Can Enzymes Be Utilized As Evidence?
In a head-on automobile collision, each driver claimed the other driver caused the accident by
falling asleep at the wheel. The two passengers, one from each car, were critically injured, yet
the drivers walked away with barely a scratch. Upon arrival at the local hospital, one of the pas-
sengers succumbed to his injuries and the accident is now a case of vehicular manslaughter. The
attending physician completed a thorough examination of the two drivers by collecting blood
and urine samples, as well as by taking their temperature. The physician saved the disposable
plastic mouthpiece and tongue depressor used during the examination, knowing that sleep de-
privation causes the level of saliva amylase to increase in humans. Students will determine the
level of saliva amylase for the two drivers to discover who was responsible for the accident.

Proteins are separated on polyacrylamide gels and are transferred (blotted) to a membrane.
