ex vivo: melanin
Melanin is the pigment responsible for skin s color and pigmentation. One of the most visible
signs of aging are age spots which are due to altered melanin distribution throughout the skin.

The testing duration with Natriance Brightener at 1% is 48 hours and 20 l is applied on skin biopsies once a day.

Arbutin at 3% and Kojic acid at 1% are used as benchmarks and applied under same conditions.

After 48 hours, melanin levels are evaluated by Fontana masson staining and quantified by color measurement with Image Pro-Analyser software.

*Efficacy above Kojic acid at 1% is considered as whitening activity in the USA and therefore falling under the requirement of an OTC drug (kojic acid CIR). See recommended use level for cosmetic application in the USA (21CFR310).

melanin index suggesting a brightening effect in vivo.

On 21 Asian volunteers (ages 22 to 29) double blind study.

The testing duration is 42 days (control at D0, D28, D42) to provide ample time to the skin to brighten. D28-D0

Melanin index (quantity of light specific to melanin wavelength absorbed by the skin) is measured with a Mexameter MX18;
Skin Luminance (real luminous intensity reflected) with a Spectrophotometer CM-2600d.

The product is applied at a dose of 4 mg/cm of product, 2 applications per day.

Statistical analysis is done using the non-parametric Wilcoxon matched pairs test.
