Method / Procedure
oven is pre-heated at 100C. Solvent beaker with extracted fat in it is taken out
using towel and moved into the oven to be dried for 30 minutes. Then, it is being
cooled using a dessicator and weighed. The formula used is

Serial Dilution
1ml of blended stock is pipette into 9ml of diluent that contain peptone water. The
mixture is then being shaken generously to ensure that it is homogeneously mixed.
After that, a subsequent transfer of same volume is done into a second tube
containing a sterile, 9ml diluent from the previous first tube. First tube labelled
with concentration of 10-1 of cells and 10-2 for the second tube. The process of
serial dilution is continued until the concentration of 10-4 is obtained.
Plating Process
0.1 ml of bacteria from each dilution is pipette onto an agar plate at the center.
Then, L-shaped glass rod or hockey stick rod is steriled by dipping it into a beaker
of ethanol and passed through a flame to burn off the alcohol. The agar surface in
the plate is spreaded evenly using the sterile glass rod. Then, the same sterilization
procedure is done again with the L-shaped glass rod. Procedure is repeated in
order to inoculate the remaining plates. Next, all of the plates are sealed and
labelled according to its number of dilution.

Incubation
Sealed plates are incubated in incubator at temperature of 30C to 35C for 48
hours. After two days, the colonies appeared on the plates are counted.

The number of bacteria between 30 to 300 is counted and data obtained is
tabulated in table as below:
