HOW TO MEASURE RATE OF TRANSPIRATION

Aim/Objective: To analyze the tempo of transpiration of a potted
plant using a potometer.

Research Question: Can the rate of transpiration of a green plant


(which is d not potted) utilizing (using) a photometer?

Hypothesis: That it is likely to measure the rate of transpiration of a


using a pototmeter. I think the result will be most likiey to be accurate
because it is easy to use the pototmeter and it will be beater.

Variables:

• Humidity
• Temperature

Procedure
1) Place the potometer in front of you on a flat surface where it is easy for
you handle it fast.
2) Handle the plastic tubing and fill it with water.
3) Make a tiny cutting of the plant, a large piece of stem, and the shoot of
the plant must be exactly in size of plastic tube, to prevent any air
bubbles in the tube, force the stem end of the cut into a container of
water right after slicing.
4) The cut is inundated , remove another 2inches or so, to make sure that
any form of bubbles is removed. Put the cutting into the tubing, and
secure it to form a seal Vaseline may help you in this.
5) Remove the green plant
6) Then place the pipette into the opposite end of the tube, and attach
the potometer against the stand.
7) Alter the relation to the height of the plant and pipette ends of the
potometer in order for the plant to be higher than the pipette side.
8) Four different biological treatments:
➢ Wind
➢ high
➢ humidity atmosphere
➢ bright light (sun light)
This would determine the baseline rate of transpiration for the room.
1) For the wind condition you can prepare a small fan in front of the
potometer blowing the air on the plant, for the humidity condition,
2) Place a large transparent polythene bag over the top of the plant
cutting, utilize the bottle spray to mist water into the transparent bag,
and secure the bag around the aril of the plant cutting.
3) For a bight light , prepare a lamp with a high watt light bulb near to the
potometer and shine mainly on the plant.

4) After all that has been prepared, wait for a few moments or so (let the
plants to transpire). Then observe the water level within the graduated
pipette. This is the starting number.
5) If you want to record the no. of water in the graduated pipette every 3
mni. for 30 mni..
