Introduction
Tobacco, the widely known drug is mainly present in
cigar, cigarette, beedi, suuff, etc. It belongs to the
family grown species are N rustica and N tobacco. One
of the harmful components of tobacco is Nicotine.
Nicotine is colorless volatile liquid. It boils at a
temperature of 246-247C and soluble in most solvents
including water. Pure nicotine is highly poisonous.It is
hygroscopic and distills over unchanged into steam. Aqueous
nicotine is strongly alkaline to litmus. It is easily soluble in

ethanol, light petroleum, ether and benzene. On exposure to air


it turns yellow and then
Brown. Nicotine is levorotatory. Nicotine is converted into
nitrogenous acid on oxidation with potassium permanganate or
chromic acid. Nicotine acid is a plant growth regulator and
vitamin. The structure of Nicotine is:-

AIM
The Aim of investigation is two-fold:1. Two extract nicotine from tobacco.
2. To determine the percentage of nicotine
in various tobacco products and make a
comparison.

PROCEDURE
About 10 grams of tobacco was grounded properly and
taken in a conical flask. Then 10 ml of conc. NaOH was
Added to it. The contents were warmed on a low flame
and stirred vigorously. This process was continued till a
strong spell of tobacco has mixed with NaOH properly,
the contents were cooled to room temperature. The
contents then were transferred to room temperature.
Then they were transferred to a separating funnel,
which had already been washed, and about 15 ml of
ether was added. Later, the contents were stirred
gently to ensure complete mixing. Some time was
allowed to elapse till the ether layer separated from
rest of the contents. Then the bottom layer of NaOH
was removed and poured back into the conical flask.
The ether was taken in a beaker and placed in a boiling
water bath to allow the ether to evaporate. The
extracted nicotine was weighed and its percentage was
calculated accordingly.

Conformation of nicotine was done by adding conc. HCL


to extracted nicotine. The quality of salt formed gave
an idea of strength of nicotine.
