OBJECTIVE :

1. Identify the type of cell respiration in the yeast.


2. Interpret the results on yeast reaction by using blue methylene as the
artificial hydrogen acceptor
INTRODUCTION :
Cellular respiration is a process of food molecules oxidation to produce
energy in ATP form: it includes aerobic and anaerobic respiration occurs in
the presence of oxygen and the oxygen act as a final electron acceptor. An
anaerobic respiration also known as a fermentation process, which occurs in
the absence of oxygen that uses an organic molecule as a final electron
acceptor.
In yeast cell, the fermentation process produce 2ATP, carbon dioxide
and the wasteful product, ethanol. During this process, NADH will donate the
atom hydrogen to keep generating of NAD and continue the fermentation
process.
MATERIAL :
Blue methylene 0.5%
Fresh yeast 9% + glucose 1%
Test tube
Water bath (38C - 45C)
Measuring cylinder 10 cm
Beaker 250 cm
Rubber stopper

The test tube A, B and C was labeled.


9 ml of fresh yeast plus 1 ml of glucose was poured into the test tube.
Placed the test tube C in a beaker of boiling water for five minutes.
10 drops of blue methylene was put into each of the test tube and
shake well.
5. Placed all the test tube in water bath for fifteen minutes. The colour in
each test tube was recorded in column x in table 3.1.
6. Placed the test tube B into a beaker of boiling water for five minutes.
7. A rubber stopper was inserted into each test tube and shake the test
tube. The colour of each test tube was recorded in column Y in table
3.1.

8. Removed the rubber stopper and placed the test tube in the water

bath for fifteen minutes. The colour of each test tube was recorded in
column Z in table 3.1.

QUESTION :
1. Explain the biochemical process that happened in the test
tubes based on your observation.
Biochemical process that happened in the test tube is redox reaction
for the test tube A is reduced process occur. For the test tube A there a
changing colour from green to colourless because it accept hydrogen.
2. Does this reaction controlled by enzyme? Give some reason.
Yes, this reaction controlled by enzyme because there no changing in
colour for the test tube C after it place in boiling water. For an enzyme
it will denature after the temperature reach it optimal value. That the
reason why the test tube C have no reaction in changing colour
because enzyme already denature.
3. Give one example of structure or substance in living cell that is
represented by blue methylene.
Methylene blue stains DNA, so one example of a structure in a living
cell that it may show by staining blue is the nucleus.
It often used to stain animal cell to make their nucleic more
observable.
4. Name the process that involves the activity demonstrated in
this experiment.
In yeast cell, glucose can be metabolized through cellular respiration.

DISCUSSION:
The sugar glucose is the main fuel source for cellular respiration.
Recall that redox reaction is simply shorthand for an oxidationreduction reaction. That mean that during cellular respiration, some of
molecule in our cellular respiration chemical will be oxidized and some
will reduced. There a lot of energy stored in the bond between the
carbon and hydrogen atom in glucose. During cellular respiration redox
reaction basically transfer this bond energy in the form of electron from
glucose to molecule called electron carries. So an electron carrier is a
basically a respiration by using an electron carries, energy harvested
from can be temporarily stored until the cell can convert energy to ATP.
Just what has happened in test tube A accept hydrogen and reduction
process occur.
For the test tube B the colour should be remain because it
already begin boiled in boiling water and the enzyme already denature
but for our experiment what we have observe is the colour change
from light green to yellow. This changing colour might happened
because of the boiling water maybe not to hot and not reach the
optimal temperature value for the enzyme, that why the enzyme is not
completely denature.
For the test tube C the colour from green remain green until the
end of experiment, this is because the test tube C is already place in
boiling water and the enzyme in that solution already denature
because of the optimal temperature already reach.
CONCLUSION :
The type of cell respiration in the Yeast is enzyme. The methylene blue
shows that test tube A is hydrogen acceptor.
