Overview
The membrane pump that attaches to the Vapur interface used with DART should be optimized upon
installation of a new unit. This document may also be referenced by a user looking to re-optimize their
membrane pump. To optimize the pump we will be running the same standard while adjust the suction
and then base the optimum setting off of the best signal response. Each pump setting should be run in
doublet.
Spot the 1-D Transmission or QuickStrip module with 3 ul of the Quinine Standard provided in
your DART kit and allow samples to dry.
Start with the needle valve one full turn out from all the way closed, as shown below.

Note: the valve has the 0 value facing up and is still in the red indicator of the needle valve
Start an acquisition on your mass spectrometer, then using the FreeRun page on your DART,
drive the first two samples through at .5mm/s and then stop the rail in between spots 2-3

Adjust the needle valve a quarter of a turn so it looks like the image below

In the same acquisition, using the FreeRun page on your DART, drive the next two samples
through at .5mm/s and then stop the rail in between spots 4-5.

In the same acquisition, using the FreeRun page on your DART, drive the next two samples
through at .5mm/s and then stop the rail at the end of the module and end the acquisition.
Adjust the needle valve a quarter of a turn so it looks like the image below.

In a separate acquisition, using the FreeRun page on your DART, drive the first two samples
through at .5mm/s and then stop the rail in between spots 2-3.

At this point, you can end the acquisition, as you should have already passed through the
optimum setting. Open up both acquisitions and pull out an extracted ion chromatogram for the
325 m/z.
Determine which of the settings gave the best signal response and reset the needle valve to that
value and lock it into place using the set screw (1.5 mm allen wrench )as shown below.
