Tissue Optics and Measurements in Biophotonics

This course presents structural and optical models of tissues with basic single and multiple scattering, with ordered
and randomly distributed scatterers. Coherent, spatially-modulated, and polarized light propagation in random and
quasi-organized tissues will be considered. It will be shown that tissue reflection and transmission, as well as
scattered intensity and polarization can be effectively controlled by changes of its structure and refractive index of
tissue components. A considerable increase of penetration depth of probing light and image contrast will be shown.
Optical monitoring of endogenous and exogenous matter diffusion within a tissue also will be demonstrated. A
considerable improvement of measurement methods and instruments, based on OCT, SHG, two-photon and confocal
microscopy, speckle and polarization-sensitive techniques, will be shown for various medical applications. These
applications include tissue and blood spectroscopy, monitoring of structure and imaging of the human skin, eye
tissues, cerebral membrane, scull, bone, cartilage, tendon, bodys interior tissues (vessel wall, esophagus, stomach),
and vessel net.

This course will enable you to:
describe coherent, spatially-modulated, and polarized light propagation in random and quasi-organized scattering
media and tissues at OC
summarize principles and schemes for basic laser instruments, such as scattering matrix meter, scanning
speckle microscope, OCT, SHG, two-photon and confocal microscopy, speckle interferometer, frequency-domain
spectrometer and tomograph, full-field speckle technique, etc.
explain mechanisms of tissue optical properties control induced by external or internal mechanical stress,
chemical agents application, temperature change, coagulation and other physical and chemical action, principles
of tumors and other tissue abnormalities contrasting
identify usage of the appropriate OCA and light scattering or coherence-domain method for optical testing, tissue
pathology study, and disease diagnostics.

INSTRUCTOR
Valery Tuchin holds the Optics and Biophotonics Chair and is a Director of Research-Educational Institute of Optics
and Biophotonics at Saratov State University, Head of Laboratory on Laser Diagnostics of Technical and Living
Systems, Inst. of Precise Mechanics and Control, RAS. His research interests include biophotonics, biomedical
optics, tissue optics, phototherapy, and optical and laser measurements in biomedicine. He has been awarded
Honored Science Worker of the Russian Federation and SPIE Fellow; he is a Vice-President of Russian Photobiology
Society. In 2007 he was awarded the SPIE Educator Award. He is a FiDiPro Professor of University of Oulu (Finland).
He has authored more than 300 peer-reviewed papers and books, including his latest, Optical Clearing of Tissues
