Earlyembryosfuelhopesforshortcuttostemcellcreation

YuryVerlinksyandhiscolleaguesattheReproductiveGeneticsInstitutein Chicago,IL,grewcellsfromfourdayoldhumanembryoscalledmorulae(see ReproductiveBioMedicineOnline www.rbmonline.com/Article/1558).Previously,stem cellshavebeengrownonlyfromblastocyststageembryos,whichareadayortwoolder andhavemorespecializedtissues. Researchersinthefieldsaythenewtechniqueisquickerandsimplerthan previousmethodsandcouldboostthesuccessrateformakingembryonicstemcelllines, whichareinshortsupply.Ittakesahugestepoutofthewholeprocess,saysPeter Braude,whostudieshumanembryonicstemcellsatGuys,KingsandSt.Thomas SchoolofMedicineinLondon. Verlinksyandhisteamattemptedtogrowstemcellsfrom46humanmorulaeleft overfrom invitrofertilization(IVF)treatment.Theysucceededinmakingeightstemcell linesasuccessrateroughlyequivalenttothatachievedusingblastocysts. Initspaper,theteampresentspreliminaryevidencethatthesestemcellsareable togenerateasimilarrepertoireofcelltypestothosegrownfromblastocysts.Theyhave alreadyusedbothtechniquestocreatealibraryofstemcelllinesfordiseaseresearch, includingmanycarryinggeneticabnormalities(seeNature429,6912004). Thenewmethodisadvantageousbecauseitavoidsthedelicatetaskofculturing embryosuntiltheygrowintoblastocysts,aperiodwhenperhapshalfofthemstop growingordie.Italsocutsoutalaboriousstepfromtheconventionaltechnique,inwhich theoutershelloftheblastocystisdestroyedtogainaccesstotheprecious2030stem cellsinside.Inthenewmethod,theentiremorulaofaround6070cellsisgrownin culture. Theadvanceisunlikelytoaffectthecontentiousglobaldebateovertheethicsof embryonicstemcellresearch,scientistssay.Opponentsofsuchresearchpointtothefact thatstemcelllinescanonlybecreatedbydestroyingahumanembryo. ButBraudenotesthatresearcherswillnowwanttotestthemoreradicalconcept ofgrowingstemcellsfromasinglecellpluckedfromamorulastageembryowithout damagingtherestofit.ThatwouldalsoraisetheprospectthatcoupleshavingIVF treatmentcouldasktohaveoneoftheirembryoscellsremovedbeforeitisimplantedin thewomb.Thecellcouldbegrownintoastemcelllineandstoredincasethechild shouldneeditfordiseasetherapyinthefuture. Becausethistechniquewouldavoiddestroyingtheentireembryo,stemcell researcherssaythatitcouldalsoservetoquellsomeoftheethicalargumentsagainstthe useofhumanembryosforresearch. Researchersmightalsousethenewmethodtogrowstemcellsfrommorulastage embryosthathavestalledintheirdevelopmentandareincapableofgrowingintobabies, suggestsstemcellresearcherJoseCibelliofMichiganStateUniversityinEastLansing. Youcouldremoveabigobstaclefromtheethicalstandpoint,hesays.
