All of these glycosphingolipid antigens are synthesized by extension from a common precursor, lactosylceramide. The likely parts of
these structures comprising the epitopes recognized by the different antibodies are highlighted. The enzymes responsible for the addi-
tion of the third sugar residues (galactosyl-, glucosaminyl-, and sialyltransferase) appear to regulate the rate-limiting step that controls
which synthetic pathway predominates. Human EC and ES cells are characterized by expression of the glycolipid antigens SSEA3 and
SSEA4, and not the lactoseries antigen SSEAl (Lex). By contrast, mouse EC and ES cells are typically characterized by their expression
of SSEAl. Mouse cells also express the globoseries Forssman antigen, which is not synthesized by human cells. Thus, despite the ap-
parent distinction between mouse and human EC and ES cells in terms of their terminal sugars that comprise the different epitopes,
the undifferentiated cells of both species do seem to share in common the expression of globoseries core structures.
