TITAN MEDIA ANAEROBIC BLOOD AGAR BASE For isolation and cultivation of Group A and B Streptococci Composition Ingredients

* Dehydrated powder, hygroscopic in nature, store in a dry place, in tightly-sealed containers below 25C and protect from direct Sunlight. Instructions for Use Dissolve 40gms in 990 ml of distilled water. Gently heat to boiling with gentle swirling and dissolve the medium completely. Sterilize by autoclaving at 15 psi (121C) for 15 minutes. Mix well. Cool to 45-500C and aseptically add rehydrated 1 vial content of Neomycin Supplement (TS 095). Mix well and add 5% v/v sterile defibrinated sheep blood. Stir properly and pour into sterile Petri plates. Appearance: Yellow colour, clear to slightly opalescent gel, Cherry red colour with the addition of 5% sterile defibrinated blood, opaque gel pH (at 25C): 7.2 0.2 Principle ANAEROBIC BLOOD AGAR BASE is used for cultivation of anaerobic bacteria especially Clostridium species. Medium contains Casein enzymatic hydrolysate and Papaic digest of Soyabean meal as a source of nitrogen, vitamins & minerals. When supplementing with blood the medium facilitates the recovery of more fastidious microorganism, such as Prevotella, Porphyromonas, and the Bacteroides fragilis group, and should facilitate pigment production of Prevotella melaninogenica. Sheep blood is added for the observation of hemolytic reactions as seen by the double zone hemoylsis of Clostridium perfringens. Sodium chloride helps to maintain the osmotic balance of the medium. Agar is the solidifying agent. Interpretation Cultural characteristics observed under anaerobic conditions after inoculating (103CFU/ml), on incubation at 35 2C for 24 - 72 hours.
