Indicator Quinoneimine dye(red-violet)

Rgt Stability 2-8 C up to expiry date o 15-25 C 2 weeks

o

Specimen Serum, plasma *glucose is stable for o 24hrs at 2-8 C if prep within 30 min after collection

Assay WL= 500 nm or Hg 546 nm OP= 1 cm T = 20-25 C o or 37

o o

Performance Linear up to glucose conc of 400 mg/dl or 22.2 mmol/L

Normal Values Serum,plasma (fasting) : 75-115 mg/dl Or 4.2-6.4 mmol/L

Notes 1.RGT contains .095% Na azide 2. icteric sera interfere with test TAG up to 2500 mg/dl, haemoglobin up to 500 mg/dl and ascorbic acid up to 20 mg/dl do not interfere 3. slight brownish sediment as RGT ages, do not swirl 1.RGT contains .05% Na azide 2. not influenced by haemoglobin values up to 150 mg/dl; bilirubin up to 40 mg/dl; ascorbate may give falsely low values at >4 mg/dl

Triglycerides

Enzymatic hydrolysis with lipases

Quinoneimine dye

2-8 C up to expiry date o 20-25 C 4 weeks

Cholesterol

Enzymatic hydrolysis and oxidation

Quinoneimine

2-8 C up to expiry date o 15-25 C 2 weeks

Serum, heparinised plasma or EDTA plama *STABILITY: o 3 days @ 2-8 C o 4 mos @ -20 C *NOTE: lipemic subs usually generate turbidity falsely elevated LCF clears up turbidity Serum, heparinised or EDTA plasma

WL= 500 nm or Hg 546 nm OP= 1 cm T = 20-35 C o or 37 C

o o

Linear up to TAG conc of 1000 mg/dl or 11.4 mmol/L

Atherosclerotic risk: Suspect: >150 mg/dl or 1.71 mmol/L Increased: >200 mg/dl or 2.28 mmol/L

WL= 500 nm or Hg 546 nm OP= 1 cm T = 20-35 C o or 37 C

o o

Linear up to cholesterol conc of 750 mg/dl or 19.3 mmol/l

Suspect: over 220 mg/dl or 5.7 mmol/l Increased: over 260 mg/dl or 6.7 mmol/l *EAS: Adults up to 30 yrs= 180 mg/dl Over 30 yrs = 200 mg/dl

1.not influenced by haemoglobin up to 200 mg/dl or bilirubin up to 5 mg/dl 2.contain 0.05% Na azide

HDL Cholesterol

Chylomicrons, VLDL,LDL are pptd by addition of phosphotungstic acid and magnesium chloride. After centrifugation supernatant contains HDL

PREC o 2-25 C up to expiry date

Serum, heparinised or EDTA plasma

WL= 500 nm or Hg 546 nm OP= 1 cm T = 20-35 C o or 37 C

WOMEN mg/dl > 65 45-65 < 45 mmol/l > 1.68 1.16 1.68 < 1.16

1.if supernatant is clear (high TAG level), dilute sample before ppt 2. high conc of ascorbic acid (. 2.5 mg/dl) will give lower values 3. haemoglobin > 100 mg/dl and bilirubin > 10 mg/dl interfere

Suspicious: 150 mg/dl or 3.9 mmol/l

Total Protein

Cupric ions react with CHON in alkaline soln to form purple complex. The absorbance of this complex is proportional to the CHON conc

2-25 C up to expiry date

Serum, heparinised or EDTA plasma *STABILITY: o 2-8 C up to 1 month 15-25 C up to 1 week

1.color rgt contains NaOJ 2. contains 0.095% Na azide as preservative 3. slight sediment as RGT ages, do not include this 4. sample blank for clear, colorless sera = 0.2 g/dl, can be ignored Sample blank must be determined for visibly haemolytic, icteric and lipemia sera by pipetting 20 ul sample to 1000 ul physiological saline and measuring against dH2O. Absorbance sample Absorbance of blank
