INTRODUCTION
Ulta structure spermatozoa studies have been
intensively examined. Information on spenmatoza ulira
ucts is needed to understand specnatoz08 biology.
hich i invaluable inthe developing of spermatozos
:zyopeeservaton methods (Lin eal, 1996) and further
‘ahuncing artificial induce breecing efforts Mati (1991)
aad been reviewed the sper morphology of around 280
“Shes ing anamisiom and ssnning leon meroseope
‘Baung isa popular catfish species curently being
produced in both Semi-intensve and intensive eu
‘stem in Malaysia, Thailan, nd Indonesia. Reproductive
Biology of baung has reported by Khan er al. (1990),
owever, information of spematozo ula stuctre of
‘ung has not been describe. The objective ofthe pedent
‘dy isto deseibe the sperm morphology ofbaung using
transmission and seaming electron micrograph imaging.
MATERIALS AND METHODS
‘The study was carried out in Eletron Microscope
Laboratory of University Sciences Malaysia, Penang,
“Malaysia. Thee male fishes weighing 58970 654-4 were
‘amaesheszed and sesificed by spinal transection. Testes
‘ere removed by dissection and perforated witha needle
snd gently squeezed ino las tobe which wa paced on
crue ie oobi the sperms.
Sperm samples were fixed in 2% glutaraldehyde and
Lopt for 48 hours. After 48 hours, sample was washed in
«acodyat buffer for one hour, end then samples were post
fixed frome hor in cacedylte bufer osmium teoxide
‘adinsed several ines in he buf. Forstanning electron
iniecoseopy (SEM), samples were attached to coverslip
viously coated with 0.1% poly-L-lysine. ARer
“ehyration through an ascending series of acetone, the
samples wert! point dei in ene point dryer, and
coated with gold in sputer costed and viewed by using
levion microscope (Leica Cambridge S-360, England),
While, for transmission elecion microscopy (TEND, the
sperm pellet obtained by gentle cenuugation were
‘delyated in acetone and embedded in ein. Thin sections
‘of samples were picked up on copper rds, doubly stained
with uranyl acetate and lad czete and exemined using
lecton microscope Philips CM 12 TEM, Holland),

DISCUSSIONS
In teleosts, the morphology ofthe spermatozoa is
simple; the hed is round or oved-shape, the middle pass
isreduced toa mitochon sing andthe gel present
‘typical "9+ 2" stratae Suquet ral, 1983) The baung
spermatozoa area typical aguasperm with aspherical
rueleus, short midpiece, and short cytoplasmic collar.
Aquaspem i alo found on other catfish suchas Atican
catfish, walking catfish, and Thai catfish. Another type is
introspem which found in intemal fertlizes 18, for
example in ocean pout (80 el, 1988.
Heid spermatozos of baun is rounded, and with no
scrosome struct tthe head. Acrosome ruta has also
absentia some species, or exsrpl ina spermatozoa
(onan Atlin, 1998) and mbot Saquet eral, 1993)
"The acrosome untion ito breakdown the icropye

‘nse rope pore which x always opening to pent
sperm rassge to feilize the egg (Amanze ane lye,
1990) Dace fertilization oceurs, the micropsle pore is
bared ty imneraus network (Ber and Avtalion, 1950),
‘The midpecesleove of baung spermatozoa is short with
single atochonda, «smile performance was fund in
‘turbot spermatozoa (Saquct era, 1993). Whereas, along
nipiee-sleve witht as nine mitbocondil was found
in eas pout (¥80 ea, 1995)
‘The are two types fish Nageiunsuniflagllt and
biflagelate, The Magellan of baung spermatozoa is
“unflagillate with axoneme formed by nine peripheral
“doubles and single pir of central microtubules (9+ 2
srt), similar pater were found in Turns thymus
and Buthynusalletteratus (Abascal ef a, 2002), and
Diplodsspuntzzo (Tad a, 2001), Usifaglat with
“9'+ 1 pattems of xoneme was found inilapia (Bere and
‘Avalos, 190), and bilagelinte with “9+ 2” setae of
‘axoneme is foand in ossn pout (a0, eta, 1995)
"Nombers of fagellum are dependent on sh species a
single Zagelium found in shsuchas iuride, Clarke,
‘Bog, Schibeidss,and Mochokidae. While biflagellum
is Tourd in Tetalurdae, Pimelodidae, Ariidae, and
‘Malaptrridae (Yao etal, 1995). this sudy, we Found
some anormal spematozoa.
‘Our observation revealed the presence of bora cell
in fos sporm (Figure 11, these abnormalities occurs
naturally and we suspected that the aboormalites of spemm
‘occur curing spermetogenesis. However, mast ofthe
‘spermatoze inthe fresh mit have anormal structure.
“Lasly we concluded hat the baung spermatozoa could
be vied into thee disinet parts; head, midpieces sleeve,
and flagellum, The bead of baung spermatozoa round
forms, and covered by eytoplasmic membrane. The
‘mdpiecesleoveis short with ar lest two mitochondria The
‘eget is uniNagoliste, withsine peripheral doublets, andMuchliin 24 n
single par of central mirotubulus (°942" patterns) of
sxoneme. Te typeof epermatozn is aquasperm.
