Background:
Yeastisasinglecellfungusthatproducescarbondioxideasabyproductofcellularrespiration.
Thereleaseofcarbondioxidecausesbreaddoughtorise.Becausetheyeastsaresmallandreproducerapidly,
yeastorganismsareusefulforstudyingvariousfactorssuchasfoodavailability,temperaturechangeandashift
inpHthatmayinfluencetherateatwhichapopulationgrows.Theoptimumtemperatureforyeastmetabolism
andyeastreproductionisbetween30and35degreesCelsiusdependingonthespeciesandprovidingthereisan
adequatefoodsupply.Thesecellsproducecarbondioxidegasasawasteproductandtheamountofcarbon
dioxideisanindicationofyeastgrowth.

b.Hypothesis.Inthespacebelow,writeahypothesisfortheproblemstatedabove.Letsmakesurethatgroups
havedecidedonwhichfactorstheywillinvestigate.BesuretowriteyourhypothesisasanIf

Iftemperaturedecrease,thenthesizeofthecarbondioxidebubblewilldecrease,becauseyeast
metabolismandyeastreproductionisbetween30and35degreesCelsius.

3.Procedure:
1. Place7mLof25%molassessolutionintoasmalltesttube.
2. Stir the yeast suspension and then place 1 mL of the yeast suspension into the same test
tube.
3. Placethetesttubeintherack.
4. Wash and rinse hands. Place a palm over the end of the small test tube and invert it five
times.
5. Carefully slide a larger tube down over the smaller tube. Quickly invert the tubes so the
mouthofthelargetubeisup.
6. Using a metric ruler measure the height of the air bubble (cm) in the smaller tube and
record.Placeinthetesttuberack.
7. Repeatsteps16fortwomoresetups.
8. Leaveonesampleat30degreesCelsiusfor24hours.
9. Leaveonesampleatroomtemperaturefor24hours.
10. Leaveonesampleinarefrigeratorfor24hours.

Ourhypothesiswascorrect,whenthetemperatureincreasestheheightofthebubblewillincrease.When
thetemperaturewas4degreescelsius,theaveragechangeintheheightofthebubblewas1.5
centimeters.Whenthetemperaturewas21degreescelsius,theaveragechangeintheheightofthe
bubblewas3.333centimeters.Andwhenthetemperaturewasat30degreescelsius,theaveragechange
intheheightofthebubblewas4.5centimeters.

Validity
:Validityisdiscussedintermsofwhatwasdoneandcouldhavebeendonetomaketheexperiment
morevalid.Howwelldidtheothergroupsconclusionsconcurwithyours?
Bymakingmoretrialstotesttheeffectoftemperaturemighthavemadetheresultsmoreprecise.Addingthe
trialswillensureareliableexperiment.Wecouldhaveincreasedordecreasedthetemperaturestogetmore
averagesontheheightofthebubble.Wecould
of
alsohavetrieddoingmorevariablesinvolvingtimelike
waitingmoredaystoseeiftheratiooftheeffectontheheightofthebubblesisstillthesame.Thiscould
improveourvaliditybecausewewilltrulybeabletoseeifourdataisspoton.

RealLifeApplication
:Howdoesthislabrelatetoreallife?
Ifwhenweremakingbread,weneedtoplacetheyeastinawarmcontrolledenvironmentin
orderfortheyeasttoadapttothewarmthofbread.Inthislabtheyeastwasinacontrolled
roomtemperaturesothatitcouldrise.
