IN VIVO STUDY ON THE EFFECT OF Mangifera indica


ABSTRACT

This research determined the anti-nephrolithic and anti-tissue necrotic activity of mango leaves
(Manifera indica) extract on male Sprague dawley rats. Ethanolic extraction method was used to get
the extract of mango leaves.

Twenty (20) male rats were divided into four groups namely the positive control, normal
control, experimental group 1 and experimental group 2. Each group received different treatment for
10 days. All were induced by 0.75% ethylene glycol and 2% ammonium chloride as nephrolithic agent
except for the normal control which remained untreated. The 300mg/kg body weight dose of mango
extract were given to experimental control 1 and the 100mg/kg body weight were given to
experimental control 2 via oral gavage. At the end of 10 days experimental period, blood was collected
at the retro orbital region and the serum was analyzed for Blood Urea Nitrogen (BUN) and Creatinine
level. The kidneys of each rat were processed for histological examination for the demonstration of
calcium oxalate deposits and tissue necrosis.

The results showed that the amount of BUN and blood creatinine level of the rats treated with
mango leaf extract significantly decreased than that of those rats treated with ethylene glycol and
ammonium chloride alone. And the histological examination results showed that there were no crystal
deposits but there were variations in the degree of necrosis among the groups. The positive control
showed the highest degree of necrosis with a P-value of <0.001, and lesser in experimental group 1 (Pvalue >0.05) and 2 (P-value <0.05). The Mango leaves was then not considered as an effective antinephrolithic agent but considered as a protective agent to the kidney.
