LAB: Movement under the Microscope Letter e Lab

Problem
How do objects look and move under a microscope?

Background
tiny, scope refers to view or look at. The compound light microscope is an instrument containing two
lenses, which magnifies, and two types of knobs to resolve (focus) the picture. Microscopes are tools used to enlarge images of small objects so as they can be studied. Micro refers to

Draw a Hypothesis
Draw a hypothesis by actually drawing what you think the letter e will look like under the microscope. Do NOT look through the microscope.

Draw letter e

Test the Hypothesis

Materials:
Microscope Slide with letter e Lab paper Pencil

Procedure (use the boxes to check off as you complete each step):
aperture (hole in the stage).

1. Turn on the microscope and place the slide on the stage so that the letter e is directly over the

2. Rotate (turn) the nosepiece so that it is on the low power objective. You will hear it click into place. 3. Turn the diaphragm so that the largest hole is directly under the aperture. Diaphragm

4. Using the coarse adjustment knob (LARGE ONE) to focus in on the letter e. 5. Keep your eye near the eyepiece and you will see the letter e come into focus. 6. Observation: Does the letter e look right side up when you look through the eyepiece? Circle: yes or no. Is the letter e facing the right way? Circle: yes or no. 7. What is the total magnification using low power? ______________ 8. Using the circle below, draw the letter e EXACTLY as you see it through the microscope.

Record and Analyze Data

Letter e Total Magnification ____________ 9. While looking through the eyepiece, slowly push the slide away from you with your thumbs. In which direction does the letter e move? Circle: Towards you or Away from you 10. Reposition the slide so that the letter e is centered over the aperture (hole in stage). Look through the eyepiece and move the slide towards you. In which direction does the letter e move? Circle: towards you or away from you 11. Reposition the slide again so that the letter e is centered over the aperture. Move the slide to the left. In which direction does the letter e move? _________________________. 12. Reposition the slide again so that the letter e is centered over the aperture. Move the slide to the right. In which direction does the letter e move? _________________________. 13. Reposition the slide once more over the aperture. Now, change from low power to medium power. You will notice the letter e is out of focus. If the letter e is out of focus, use the coarse adjustment knob to re-focus the letter e.

14. Looking through the microscope, draw the letter e as you see it. Calculate total magnification under medium power and write it on the line below.

Letter e Total magnification _____________

15. Study each of the drawings shown below. Each drawing represents how an object looks under the microscope. Describe how you would move each slide in order to center the specimens under the microscope. The first one is done for you.

Conclusion: Write in complete sentences.

1. Explain what happened to the letter e when you place it right side up under the microscope. __________________________________________________________________________________ __________________________________________________________________________________ __________________________________________________________________________________ 2. Explain what happened to the letter e when you moved it to the right or to the left of the stage. (Hint: does the letter e travel in the direction you moved it while looking through the eyepiece?) __________________________________________________________________________________ __________________________________________________________________________________ __________________________________________________________________________________ 3. Describe the changes in appearance of the letter e when you moved the objective lenses from low power to medium power. (Hint: does the letter e get larger or smaller?). __________________________________________________________________________________ __________________________________________________________________________________ __________________________________________________________________________________ 4. Do your results accept or reject your hypothesis (the first drawing of the letter e? __________________________________________________________________________________ __________________________________________________________________________________ __________________________________________________________________________________
